| Objective:In this thesis,the variation tendency and expression level of mucin2(MUC2)and Villin in the tissue of human colon adenocarcinoma,colon adenoma,and reasonable control has been studied.The clinical significance in the occurrence and development of the colon adenocarcinoma are also explored.Methods:In this paper,3 types of different colonic tissues,including 10 nomal colonic tissues,16 colonic adenoma tissues and 40 colon adenocarcinoma tissues are selected as experimental samples.By using Real-timePCR and SP immunohisto-chemical methods,the expressions of MUC2 and Villin in these kinds of colonic tissues are detected.Meanwhile,the relationships of MUC2 and Villin to clinic pathologic features are analyzed with the clinical pathological data.Results:1.The mRNA and protein expression of MUC2 in the tissue of normal control,colon adenoma and colon adenocarcinoma both are in gradually decreasing trend;Compared with the colonic adenoma group,the colon adenocarcinoma group is statistically different(P<0.05);and compared with the normal colonic tissue group,the colon adenocarcinoma group is also significantly different(P<0.05);MUC2 in colon adenocarcinoma is not related to gender,age and location of tumor(P>0.05);but it really is related to the depth of invasion of tumor,lymph node and organ metastasis(P<0.05);the greater the depth of tumor invasion with lymph and organ metastasis is,the weaker the expression is(P<0.05)2.The mRNA and protein expression of Villin in the tissue of normal control,colon adenoma and colon adenocarcinoma both are in gradually decreasing trend;Compared with the colonic adenoma group,the colon adenocarcinoma group is statistically different(P<0.05);and compared with the normal colonic tissue group,the colon adenocarcinoma group is also significantly different(P<0.05);Villin in colon adenocarcinoma is not related to gender,age and location of tumor(P>0.05);it is also related to the depth of invasion of tumor,lymph node and organ metastasis(P<0.05);the greater the depth of tumor invasion with lymph and organ metastasis is,the weaker the expression is(P<0.05);3.There is no significant correlation between the mRNA and protein expression of MUC2 and Villin in the tissue of normal control,colon adenoma and colon adenocarcinoma(P>0.05).Conclusion:1.The expression of MUC2 and Villin decrease gradually during the evolution process from normal control to colon adenoma and to colon adenocarcinoma,which indicates that they are both correlated with the occurrence and development of colorectal adenocarcinoma.2.The expression of MUC2 has a negative correlation with the lymphatic metastases,distant metastases and infiltration depth of colorectal carcinoma(P<0.05),which shows that MUC2 may inhibit the progress of colon cancer and lymph node and distant metastasis.And it may indirectly reflect the progress of the tumor;3.The expression of Villin has a negative correlation with the lymphatic metastases,distant metastases and infiltration depth of colorectal carcinoma(P<0.05),which shows that Villin may inhibit the progress of colon cancer and lymph node and distant metastasis.And it may indirectly reflect the progress of the tumor. |
