Expression Of Villin And MUC2 In Carcinogenesis And Progession Of Colon Carcinoma And Their Clinical Significance

Objective: To study the expression of villin and MUC2 in the stages of carcinogenesis and progession in colon carcinoma and to find out the relationship between the expression level and clinico-pathological characteristics. and between the expression of them and the tumorigenesis of colon adenomas.and explore their relationship.Methods:Detecting villin and MUC2 proteins by using PicTureTM immunohistochemical staining technique in 40 resected colon mucosa ,20 colon adenoma (13 lower-moderate dysplasia and 7 severe dysplasia.)and 40 colon adenocarcinoma mucosa and 40 the matching peri-carcinoma mucosa.Results:(1)The results showed that villin positive staining was found in cell membrane with brush border microvilli and in the apical cytoplasm,adjacent to the villus partly,but negative in goblet cell. There was different degree heterogenecity in the expression of villlin in colon carcinoma and colon adenoma.For the resected colon mucosa ,colon adenoma and colon adenocarcinoma ,the positive rates of expression of villin were 90.0%,75.0%,50.0% respectively,and the difference among these groups were significant(P<0.05).The difference between resected colon mucosa and lower-moderate dysplasia mucosa were not significant(P>0.05).but the difference were significant between resected colon mucosa and severe dysplasia mucosa (P<0.05). For adenocarcinoma and heavy grade dysplasia of adenomas ,there were no significant difference in villin protein expression between them.(P>0.05).The difference between groups of adenoma were significant(P<0.05). For resected colon,mucosa,peri-carcinoma mucosa and colon adenocarcinoama,there were also statistical among the three groups(P<0.05). )The expression of villin was negative correlated to the depth of invasion(P<0.05),but unrelated to differentiation and lymph node metastasis(P>0.05)(2) The results showed that MUC2 positive staining was found in the perinuclear cytoplasm of cells with a goblet cell type theca and with apical mucous droplets . There was different degree heterogenecity in the expression of MUC2 in colon carcinoma and colon adenoma.For the resected colon mucosa ,colon adenoma and colon adenocarcinoma ,the positive rates of expression of MUC2 were 95.0%,80.0%,55.0% respectively The difference among these groups were significant(P<0.05).
