| Objective:To establish the microscopic identification of mulberry branches and thin layer identification of multi-indicator components and determining the content of multiple indicators,complete the examination of routine items by using plenty of methods and techniques.On this basis,to be written a draft for improving the quality standards of Xinjiang mulberry branches.In addition,to study on the hypoglycemic activity about mulberry branches of water-soluble alkaloids(MASA)by using of animal experiments.Methods:1.To identify the mulberry branch powder by using fluorescence inverted microscope.2.The indicators of mulberry branch-glycoside A,rutin,scopolamine,chlorogenic acid,scopolamine,allosin,resveratrol,quercitrin,1-deoxynohalide(DNJ),γ-aminobutyric acid(GABA)were identified by using thin-layer and sub-microscopic chromatography.3.The water,total ash,acid-insoluble ash,alcohol-soluble extract and other routine items to be checked for ten batches of mulberry branches.4.The content of mulberry glycosides A,chlorogenic acid,scopolamine,resveratrol,selinin,DNJ and GABA ware determined by HPLC-UV.5.MASA was isolated and administered intragastrically to alloxan-induced diabetic mice.Body weight,blood glucose,organ index,liver and muscle glycogen and other indicators were measured.Results:1.Ramulus mori has crystal thick wall cells,calcium oxalate crystal,stone cells,cork cells,wood ray cells,wood fibers and different types of catheters and other features by microscopic identification.2.The optimum conditions of thin layer chromatography for identification of the different indicators:in the silica gel G plate,with the n-butanol-glacial-acetic acid-water(90:24:24)as the developing agent,chlorine-tolidine as the chromogenic agent,to view under the white light,DNJ from mulberry branches can be better separated and identificaed;in the silica gel G plate;with n-butanol-glacial acetic acid-water(8:4.4:2)as developing solvent,0.2%ninhydrin solution as developer,heated to 105℃ color appears,to view in the white light,GABA from mulberry branches for can be better separated and identified;in the silica gel G plate,with chloroform-ethyl acetate-formic acid(44:30:27)as the system,5%AlCl3 ethanol solution as the reagent,under the temperature of 105℃heating 5 min,under 366 nm to view,which can be separated and identified the mulberry glycosides A,rutin,scopolamine,chlorogenic acid,scopolamine,sesamin;in the polyamide film,by using 75%SDS-n-butanol-heptane(27:63:10)microemulsion-formic acid(9:1)as developing solvent and 1%AlCl3 ethanol solution as developer,under 366 nm to view,resveratrol,sencin,quercitrin,scopolamine,chlorogenic acid,rutin,mulberry glycosides A,scopolamine and other indicators of 8 components can be separated and identified from mulberry branch.3.Ten batches of mulberry moisture content of 5.70%~8.02%,total ash 2.60%-4.72%,acid insoluble ash 0.47%~1.47%,alcohol-soluble extract 1.65%~2.71%.4.Ten batches of Mulberry glycosides A not less than 1.33 mg/g,chlorogenic acid not less than 0.39 mg/g,scopolamine not less than 0.37 mg/g,resveratrol not less than 0.05 mg/g and not less than 0.22 mg/g for suncin,not less than 50 μg/g for DNJ and not less than 11.0 pg/g for GABA.5.MASA can alleviate the symptoms of weight loss in diabetic mice,improve tissue organs such as hyperplasia,congestion and atrophy and other pathological changes,reduced the blood glucose,and promote glycogen and muscle glycogen synthesis.Conclusion:1.The study is based on the Chinese Pharmacopoeia quality standards as a guiding principle,the establishment of the Xinjiang Mulberry identification,inspection,determination of the content,finally,writing the improvement draft quality standards in the basis.2.Preliminary identified the water-soluble alkaloids of Xinjiang mulberry can reduce the blood glucose levels. |
