Objective:To observe the effect of periodic acupoint catgut implantation on the quanity of serum estrogen(E2),progesterone(P)and estrogen receptor(ER)mRNA in rats model of Hyperplasia of mammary gland,and to explore the mechanism of the treatment of Hyperplasia of mammary gland by periodic acupoint catgut implantation.Methods:32 Female SD Rats were randomly divided into normal control group and model control group.The rats from model control group were intramuscular injected of estrogen daily for 25d,then were intramuscular injected of progesterone daily for 5d.After 1 months of modeling,observing the appearance,height of nipple,and Histopathology of the rat’s breast were determined,after confirming the success of the model,the rats from model control group were randomly divided into the model control group,periodic acupoint catgut embedding group,and the medicine group,8 rats in each group.After observing the sexual cycle,periodic acupoint catgut embedding group selected Tanzhong and Wuyi,and according to the different stages of sex cycle to select the catgut and embed line,each sex cycle embeded 1 times,1 times was for a course,one treatment included 3 courses.Medicine Group were gavaged with TAM(1.8mg/kg/day),1 time per day,the course of treatment was the same as periodic acupoint catgut embedding group.The model control group was daily headed and grabbed,then were gavaged with NS,1 time per day,the course of treatment was the same as periodic acupoint catgut embedding group.The normal control group was daily headed and grabbed,without any treatment.The diameter and height of the 2nd and 3rd pairs of nipples were measured at the end of the treatment and the appearance of the rats’ breast s were observed.After the treatment,all rats were killed,the serum E2 and progesterone levels of the rats were measured by Radioimmunoassay,and the expression of estrogen receptor gene in breast tissue was detected by Q-PCR.Results:1.Rat nipple height and diameter:Compared with the model group,the second pair of nipple height and diameter were significantly decreased in periodic acupoint catgut embedding group and medicine group,and there were significant differences between the periodic acupoint catgut embedding group and the model control group(P<0.01),and the medicine group had significant difference(P<0.05)compared with model control group.2.Histopathology of rats’ breast:Compared with the model control group,the glandular vesicles of the mammary gland in periodic acupoint catgut embedding group and medicine group were reduced,the extent of mammary duct and acinar dilatation decreased,and the hyperplasia of mammary gland decreased obviously.3.The results of Radioimmunoassay in detecting serum E2 and progesterone levels in rats showed that:compared with the blank group,the serum concentration of E2 in the model group increased significantly,and the serum P content was significantly decreased,and there were significant differences(P<0.01),Compared with the model control group,the serum content of E2 in the rats was significantly decreased and the serum p content increased significantly,but the serum content of E2 in medicine group was significantly decreased,and the serum p content increased significantly,all of them had significant difference(P<0.05).Compared with medicine group,there was no significant difference in serum E2 and P levels in the rats with periodic acupoint catgut embedding group(P>0.05).4.The expression of estrogen receptor mRNA in the mammary gland of rat was detected by Qtime fluorescence quantitative PCR,and the expression of estrogen receptor mRNA in the rat mammary gland was significantly increased in the model control group compared with the normal control group(P<0.01).Compared with the model control group,the estrogen receptor mRNA in the breast tissue of the group and the medicine group were significantly decreased,there was significant difference(P<0.01),and compared with medicine group,the expression of estrogen receptor mRNA in the mammary gland of rats was significantly decreased,and there was significant difference(P<0.01).Conclusion:The method of periodic acupoint catgut embedding can inhibit and improve hyperplasia of mammary gland,and the effect of regulating serum sex hormone level is comparable to that of TAM,and the effect of regulating estrogen receptor mRNA gene expression of mammary gland is better than TAM.
