Study Background and Objective: Hyperplasia of Mammary Gland(HMG) is anon-inflammatory, non-neoplastic diseases, estrogen and progesterone imbalance led tothe substance of breast hyperplasia and subinvolution, it is the benign proliferativehyperplasia of the breast in the real terms.It is a common disease in the20to50-year-old female breast disease. For its high incidence and some may be cancerous,modern medicine commonly used in hormone therapy, it can play a therapeutic role ingiven period to relieve symptoms, but meanwhile, it can cause side effects such asendocrine disorders. Traditional Chinese Medicine has accumulated a wealth ofexperience in the treatment of HMG, by using the overall concept to diagnosis andtreatment. A large number of clinical reports suggest that as one of the TCM treatments,acupuncture can inhibit and reverse the pathological change of the HMG, overallregulate the endocrine, to some extent, can be effective in preventing cancer, and hasgood long-term efficacy. This study was designed to observe the effect of catgutimplantation at acupoint on the quantity of E2and expression of PRã€PCNA in ratsmodel of HMG, and histological changes in breast tissue, to explore the possiblemechanism of acupuncture treating HMG. And by setting the confrontation group, toinvestigate the preventive effect of acupuncture on HMG treatment.Material and Method:60SD rats (all females are not pregnant) were randomlydivided into6groups, normal control group, model control group, acupuncture group oftwo courses, confrontation group of two courses, acupoint catgut embedding groups of1course and2courses,10rats in each group. Modeling method: establish HMG animalexperimental model by exogenous injection of estrdiol benzoate and progensterone. Modeling for1month, randomly selected one from the model group to dothepathological examination of breast tissue. After the modeling was recognized toconduct the intervention study. The confrontation group should begin the catgutembedding therapy and modeling at the same time.After the intervention, measure the height and diameter of the second pair of rats’nipple. Blood by remove the eyeball method, separate the serum, then test the quantityof E2by using the electrochemical immunoluminometric method. After anesthetized,cut the second pair of rats’ nipple and fixed, embedded and sectioned, then use theoptical microscope to observe the breast histopathology change. Detected the expressionof progesterone receptor PR and cell proliferation-related protein PCNA by usingImmunohistochemical methods, and do the qualitative and quantitative analysis of theexperimental results.All the experimental data was statistical analysed by SPSS19.0statistical softwarefor windows, take P<0.05as statistically significant differences, P<0.01as extremelystatistically significant differences.Results: Compared with normal control group, model serum E2level of controlgroup was extremely significantly increased, PR and PCNA protein expression wasextremely significantly increased in breast tissue (P<0.01), rats’ nipple and the tissuesamples significantly changed, the histopathology was also quite different. Comparedwith the model group, E2levels of acupuncture and different acupoint catgutembedding therapy groups decreased in varying degrees, and the expression of PRã€PCNA in breast tissue weakened in varying degrees.Pathological changes in breasttissue, nipple appearance were improved in varying degrees.Conclusion: Abnormal changes in serum E2levels and abnormal expression ofPRã€PCNA protein in breast tissue may be part of important inherent factor of HMG.Catgut implantation at acupoint and acupuncture treatment can reduce the serum E2levels, regulate the dynamic balance of hormone levels to restore its normal adjustmentin the pituitary-ovarian-endocrine axis, down the expression of PR and PCNA in breasttissue, reverse regulate the cell proliferation, inhibit the abnormal cell proliferation,block or even reverse the pathological changes of the rats mammary gland hyperplasia.So preliminary infer that this may be part of the mechanism of actgut implantation atacupoint and acupuncture treat the HMG. |
