ObjectiveThis study aimed to explore tumor suppressor gene RNF180(ring finger protein 180)protein expression in renal cell carcinoma tissues and explore its expression in paraffin embedded specimens of significance in renal clear cell carcinoma and its relationship with clinical pathological characteristics of renal cell carcinoma,providing the basis for targeted therapy.Methods1.Through the detection of tumor suppressor gene RNF180(ring finger protein 180)protein in renal cell carcinoma collected fresh tissues of the First Affiliated Hospital of Jinzhou Medical University from January 2012 to December 2015,225 cases of paraffin embedded specimens and paired patients with renal cell carcinoma after distal normal tissue with HE staining that representative structure,prepared by tissue microarray seven rows and ten columns using tissue film mechanism,expression of RNF180 protein was detected by immunohistochemical method,the relationship between clinicopathological characteristics and SPSS analysis of RNF180 protein expression and renal cell carcinoma.2.60 cases were collected from April 2016 to Spetember 2017 after diagnosis for patients with renal cell carcinoma,surgery fresh tissue specimens and corresponding normal fresh tissue,cryopreservation,tissue protein extraction,expression of RNF180 protein by Western water blotting assay in tissue level,and use ImageJ software to measure the expression of RNF180 protein in gray value and,using GraphPad Prism6 analysis of RNF180 protein expression in clear cell renal cell carcinoma and adjacent.Results1.The expression of RNF180 protein in the fresh tissue was significantly lower than that in the normal tissue(P<0.05).2.The results of immunohistochemistry showed that the positive expression of RNF180 protein in renal clear cell carcinoma was 48.9%(110/225)was significantly lower than that in normal tissues 75.5%(170/225,P=0.000);The expression of RNF180 protein in renal clear cell carcinoma was not related to age,sex,TNM staging and tumor size of the patients(P values were greater than 0.05).3.Kaplan-Meier analysis showed that the expression of RNF180 protein was positively correlated with the good prognosis of renal clear cell carcinoma(P=0.001).4.Multivariate Cox regression analysis showed that the risk coefficient of tumor diameter less than 4cm is tumor diameter greater than 4cm 16.1%(P=0.000),TNM,beside the risk coefficient of stage I is the other 17.7%(P=0.000),while the positive expression of RNF180 protein expression on prognosis than patients with a negative 3.175 times(P=0.001),both of the three as the independent prognostic factors in renal cell carcinoma,but not with gender and age and prognosis.ConclusionsRNF180 inhibits the development of renal clear cell carcinoma and is a poor prognostic indicator of renal clear cell carcinoma,and is expected to be a new target for molecular targeting therapy.The expression of RNF180 protein in fresh tissue of renal cell carcinoma and a large number of tissues in paraffin embedded tissues were lower than normal tissues;It may be involved in the tumorigenesis inhibition of renal cell carcinoma,but it also is a good prognostic indicator of renal cell carcinoma,and is expected to become a new target for molecular therapy.
