Objective: In this study,we detect Sprouty-related proteins with EVH1 domain(SPRED2)and microtubule-associated protein light chain 3 Ⅱ(LC3Ⅱ)expression in the clear cell renal cell carcinoma(cc RCC)tissues,and compare their expression levels with patients’ characteristics for the sake of their clinical significance.Methods: Total cases of 10 normal renal paraffin-embedded tissues,50 ccRCC paraffin-embedded tissues and 35 adjacent paraffin-embedded tissues were collected from patients with postoperatively pathological diagnosis.According to immunohistochemistry(IHC),we detected SPRED2 and LC3 Ⅱ expression in above tissues,and then investigated their clinical significance.Results:1.SPRED2 was located in the cytoplasm and cell membrane of ccRCC,mainly in the cell membrane.SPRED2 expressed higher in cc RCC tissues and adjacent tissues,instead of normal renal tissues(P<0.05).Besides,the expression levels of SPRED2 in cc RCC tissues(6.86±2.16)were obviously higher than in adjacent tissues(4.16±1.58)(P<0.05).the expression of SPRED2 in cc RCC tissues negatively associated with patient’ clinical stage and TNM stage(P=0.02 and P=0.01,respectively),however had no correlation with age,gender,lymph node metastasis and distant metastasis(all P>0.05).The expression of SPRED2 in adjacent tissues negatively associated with patient’ clinical stage and TNM stage(P=0.005 and P=0.043,respectively),but had no correlation with age,gender,and distant metastasis(all P>0.05).2.LC3 Ⅱ was located in the cytoplasm and cell membrane,mainly in the cell membrane.LC3 Ⅱ strongly expressed in cc RCC tissues and adjacent tissues.LC3 Ⅱ expression levels was significantly higher in adjacent tissues(6.63±2.12)instead of cc RCC tissues(4.20±1.73)(P<0.05).The expression levels of LC3 Ⅱ in both cc RCC tissues and adjacent tissues were higher in normal renal tissues(all P<0.05).However,the expression of LC3Ⅱ had no correlation with age,gender,lymph node metastasis and distant metastasis(all P>0.05).Conclusion:1.SPRED2 expression was increased in ccRCC tissues and adjacent tissues.SPRED2 expressed higher in adjacent tissues compared with cc RCC tissues or normal renal tissues.2.The expression of SPRED2 in cc RCC tissues negatively associated with patient’clinical stage and TNM stage,however had no correlation with age,gender,lymph node metastasis and distant metastasis.3.LC3 Ⅱ strongly expressed in ccRCC tissues and adjacent tissues.the expression of LC3 Ⅱ had no correlation with age,gender,lymph node metastasis and distant metastasis.
