| Purpose The incidence of cleft lip and palate is high in China,with which the alveolar cleft often coexists.While the alveolar cleft has both defects and deformities of bone and soft tissue,which places a great burden on patients and even society.This study intends to explore a safer,more reliable,and more convenient treatment to repair alveolar cleft or create conditions for successful bone grafting.Methods Rabbit adipose-derived stem cells were isolated and cultured in vitro.Then osteogenic induction was performed using osteogenic medium.The biological characteristics of adipose-derived stem cells were detected by flow cytometry,MTT assay,ALP assay,and alizarin red staining.In vitro studies,the characteristics of cell scaffold complexes were detected by CCK-8 assay,cell adhesion assay,HE staining,and scanning electron microscopy.Finally,cell scaffold complexes repairing the alveolar cleft in the body were examined by observation,X-ray detection,scanning electron microscopy,and western blot.Results Rabbit adipose-derived stem cells were successfully isolated and cultured in vitro,and they were successfully induced into bone cells.The biological properties of the cell scaffold complexes were stable and could repair the rabbit alveolar bone defects.Conclusions Although bone grafting is currently the main method for alveolar clefts,it is not suitable for patients with large fissures or bone graft failures.In addition,the iliac bone grafting will induce large wound and many complications.Therefore,our study figures out that using autologous adipose-derived stem cells combined with polylactic acid scaffolds to repair bone defects is a safe and reliable treatment. |
