Objective: Rat traumatic skin ulcer model by surgical method and treated with Kui Yu Ling ointment,observated the rate of wound healing and relevant factors in the process of wound healing,and explore the mechanism of Kui Yu Ling ointment promote of traumatic skin ulcer healing in rats.Method: The skin ulcer models in 72 male SD rats were established surgically and randomly divided into the control group,the MEBO group,and the Kui Yu Ling group,24 rats in each group.On the 3rd,7th,14 th days of treatment of wound area measurement and calculation of ulcer healing rate;observation of granulation tissue and capillaries HE staining;immunohistochemistry was used to detect Ang-1,Ang-2 and wound Tie-2 protein expression and calculate the expression rate of positive cells;Western Blotting method to detect Ang-1,Ang-2 and the expression level of Tie-2 protein content;The concentration of interleukin-1β(IL-1β)in the serum was detected by enzyme linked immunosorption(ELISA).Result: 1.Wound healing rate on 3rd day of treatment,the healing rate of rats in each group,the difference with no statistically significant(P>0.05);on the 7th,14 th days,MEBO group,Kui Yu Ling group wound healing rate were higher than the control group,the differences were statistically significant(P<0.01),MEBO group ulcer healing rate is quite similar with Kui Yu Ling group,the difference with no statistically significant(P>0.05).2.HE staining results.On the 3rd day,under the microscope,there were obvious inflammatory reaction and extravasation of red blood cells,compared with control group,MEBO group,Kui Yu Ling group red cell exudation is light,a small amount of new small blood vessels.On the 7th day,red blood cell exudation and a large number of neutrophils infiltrated in the control group.Compared with the control group,the number of neutrophils in the MEBO group and the Kui Yu Ling group increased significantly,and the growth of new small vessels were more obvious,and the growth of fibroblasts were vigorous.The growth of granulation tissue in the Kui Yu Ling group was more vigorous than that in the MEBO group.On the 14 th day,the granulation tissue of each group was rich,and the granulation tissue arranged more closely and orderly in the MEBO group and Kui Yu Ling group than that in the control group,and the neovascularization gradually transformed into small motion / vein.3.The positive cell rate and the localization expression of Ang-1,Ang-2 and Tie-2 protein labeled by immunohistochemistry.Ang-1 location is expressed around the vessels,and is brown and yellow to russet.Ang-2 is expressed in interstitial spaces,and the color is russet.Tie-2 is expressed in vascular endothelial cells with brown or brown and yellow color.The positive expression rate of Ang-1 on the 3rd,7th,14 th days were lower in the control group than that in the MEBO group and Kui Yu Ling group,the differences were statistically significant(P<0.01),and the MEBO group was lower than the Kui Yu Ling group,the difference had statistically significant(P<0.01).The positive expression rate of Ang-2 on the 3rd,7th,14 th days were higher in the control group than in the MEBO group and Kui Yu Ling group,the differences were statistically significant(P<0.05),On the 3rd day,the MEBO group was higher than the Kui Yu Ling group,and the difference was statistically significant(P<0.01).The positive expression rate of Tie-2 on the 3rd,14 th days were lower in the control group than that in the MEBO and Kui Yu Ling group,the differences were statistically significant(P<0.05).The MEBO group was lower than the Kui Yu Ling group,the difference was statistically significant(P<0.05).On the 7th day,control group and the MEBO group were all lower than those of the Kui Yu Ling group,the differences were statistically significant(P<0.05).4.Ang-1,Ang-2,Tie-2 protein expression level.The expression level of Ang-1 in Kui Yu Ling group was higher than that of MEBO group and control group on the 3rd,7th,14 th days of protein,the differences were statistically significant(P<0.01),MEBO group was higher than control group(P<0.01);the expression level of Ang-2 protein in control group was higher than that of the 3rd day Kui Yu Ling group and MEBO group,the differences were statistically significant(P<0.01),Kui Yu Ling group was higher than that of MEBO group(P<0.01).On the 7th day,control group was higher than the MEBO group and the Kui Yu Ling group,the differences were statistically significant(P<0.01),and the MEBO group was higher than the Kui Yu Ling group,and the difference was statistically significant(P<0.01).14 th day,the control group was higher than that of MEBO group,Kui Yu Ling group,the differences were statistically significant(P<0.01),MEBO group was higher than that of Kui Yu Ling group,the difference was statistically significant(P<0.01).The expression level of Tie-2 in control group was higher than that of MEBO group,Kui Yu Ling group on the 3rd,7th,14 th days of protein,the differences were statistically significant(P<0.01),MEBO group was higher than that of Kui Yu Ling group,the difference was statistically significant(P<0.01).5.the content of serum IL-1β.On the first day of treatment,the level of serum IL-1β were similar in each group(P>0.05).On the 3rd,7th days,Kui Yu Ling group and MEBO group serum IL-1β levels were lower than the control group,the differences were statistically significant(P<0.01),the Kui Yu Ling group was lower than that of MEBO group(P>0.05);On the 14 th day,Kui Yu Ling group lower than that in the control group,there was significant difference(P<0.05).The content of Kui Yu Ling group was lower than that of the MEBO group,and the MEBO group was lower than the control group,and the difference with no statistically significant(P>0.05).Conclusion: 1.Kui Yu Ling ointment can promote the growth of capillaries in the granulation tissue of the skin ulcer rats,make the granulation tissue grow well and improve the healing rate of the wound.2.Kui Yu Ling ointment can reduce the inflammatory reaction of skin ulcer,and create favorable conditions for wound healing.Its mechanism may be related to decreasing the content of IL-1β in serum.3.Kui Yu Ling ointment can increase the expression level of Ang-1 and Tie-2 protein in cells,reduce the expression level of Ang-2,thereby promoting the growth of vascular endothelial cells and angiogenesis,accelerating the healing speed of wounds. |
