| Purpose: The experimental chronic heart failure rat model was established by intraperitoneal injection of adriamycin,observation of Shenfu Qiangxin Decoction on chronic heart failure(CHF)rats serum brain natriuretic peptide(BNP)level and left ventricular ejection fraction(LVEF)to investigate the effects of Shenfu Qiangxin Decoction in treating chronic heart failure mechanism,for Shenfu Qiangxin Decoction application provide reliable experimental basis for the clinical.Method:Take the SPF level,weight of male SD rats 180-220 g 40.Randomly selected 10 SD rats as the control group,the remaining 30 heart failure model group by intraperitoneal injection once a week.Adriamycin(4mg/kg),for 6 weeks to establish the rat model of chronic heart failure after.6 weeks,two groups randomly a total of 10,detected by echocardiography,serum BNP level,if the serum BNP of rats in model group and heart failure increased,showed by lvef60%,and the rats diets,hair color,abnormal mental state,that was constructed successfully.The successful model after the model rats were randomly divided into three groups: Chinese medicine group,western medicine group,model group,control group of normal rats.More than four of 10 rats in each group.The Chinese medicine group with the preparation of shenfuqiangxin Decoction(2.46g/kg/d)treatment,western medicine group was given captopril(2.25mg/kg/d)feeding,blank group and model group were perfused with The volume of distilled water,the four groups were treated once a day for 4 weeks.After the prognosis of the drug,the LVEF was measured by Doppler color Doppler ultrasound.The abdominal artery blood was measured by BNP,and the pathological changes of myocardium were observed under light microscope,then the records were recorded and analyzed statistically.Result: 1.Results of serum BNP of rats in each group: compared with the blank group,model group,Chinese medicine group,western medicine group serum BNP levels were kincreased,the difference was statistically significant(P < 0.01);compared with the model group,Chinese medicine group,the serum level of BNP rats decreased,the difference was statistically statistically significant(P < 0.01);traditional Chinese medicine group andWestern medicine group compared with no significant difference(P > 0.05).2.Groups of rats: comparison of LVEF results with the control group,model group,Chinese medicine group,western medicine group rats LVEF value decreased significantly,the difference was statistically significant(P < 0.01).Compared with the model group,Chinese medicine group,western medicine group rats of LVEF increases,the difference was statistically significant(P < 0.01);traditional Chinese medicine group and Western medicine group compared with no significant difference(P > 0.05).3.Rats pathological results:The pathological sections of the blank group showed that the size of the rat cardiomyocytes was normal,the cells were homogeneous,the cytoplasm was homogeneous and the fibers;The pathological sections of the model group showed that the rat cardiomyocytes were significantly larger,the disorder was arranged,the cytoplasm was stained,the myocardial fiber rupture.And there was a significant difference between the blank group and the blank group;The cardiomyocytes of the traditional Chinese medicine group and the western medicine group were slightly enlarged,the myocardium was arranged neatly,the cytoplasm was relatively uniform.There was no significant difference between the two.Conclusion:1.By intraperitoneal injection of adriamycin in rats,detected by echocardiography and serum BNP level in rats,mental state,activity,diet,hair growth,body weight changes,proved that this modeling method is feasible and can be used for experimental study.2.Shenfu Qiangxin Decoction can reduce the level of serum BNP in rats with chronic heart failure,and the effect is similar to that of Western medicine group.3.Shenfuqiangxin decoction can improve the level of LVEF in rats with chronic heart failure and improve heart function,and the effect is similar to that of Western medicine group.4.Shenfu Qiangxin Decoction can relieve the pathological damage of myocardial cells in rats with chronic heart failure.5.Chronic heart failure TCM pathogenesis of heart qi with Yang,causing blood stasis,water retention.The clinical with Yiqi Wenyang Huayu Lishui,as the main treatment. |
