Objective:To observe the effect of Qiangxin Capsule on Ca2+-Mg2+-ATPase activity in rats with heart failure,and to explore the possible mechanism of Qianxin Capsule in delaying myocardial energy metabolism during heart failure.Methods:100 male Sprague-Dawley rats were randomly selected 10 as normal group(KG).The remaining 90 rats were treated with intraperitoneally injection with Adriamycin(ADR)to establish a model of rat with heart failure.After the successful establishment of the model,50 heart failure rats were established.The successful rats with this model were randomly divided into 5 groups: model group(MG),trimetazidine hydrochloride group(YG),Qiangxin capsule high-dose group(QG),Qiangxin capsule middle-dose group(QZ)and Qiangxin capsule low-dose group(QD).Rats were given continuous administration 4 weeks.Then the activities of Ca2+-Mg2+-ATPase,left ventricular ejection fraction,and heart weight index of myocardial cells in heart failure rats were measured.Results:(1)Comparison of myocardial mitochondrial Ca-Mg-ATPase activity: Compared with the normal group(KG),the other groups all decreased to varying degrees.The rat of the model group(MG),the Qiangxin capsule middle-dose group(QZ)and the Qiangxin capsule high-dose group(QG)were lower than the normal group(KG),the difference was statistically significant(P<0.01);The rats of the normal group(KG),the trimetazidine hydrochloride group(YG)and the Qiangxin capsule high-dose group(QG)were higher than the model group(MG),the difference was statistically significant(P<0.01).(2)The effects of left ventricular ejection fraction on Echocardiographic(LVEF): Compared with the normal group(KG),the LVEF level of the other groups were significantly reduced.The level of the model group(MG),the Qiangxin capsule middle-dose group(QZ)and the Qiangxin capsule low-dose group(QD)were significantly lower than the normal group(KG),the difference was statistically significant(P<0.01);The level of the normal group(KG),the trimetazidine hydrochloride group(YG),the Qiangxin capsule high-dose group(QG)and the Qiangxin capsule middle-dose group(QZ)were higher than the model group(MG),the difference was statistically significant(P<0.01 or P<0.05);(3)Comparison of heart weight index(HWI):Compared with the normal group(KG),the index of the other groups increased to some extent.The index of the model group(MG),the Qiangxin capsule middle-dose group(QZ)and the Qiangxin capsule low-dose group(QD)were lower than the normal group(KG),the difference was statistically significant(P<0.01 or P<0.05);The index of the trimetazidine hydrochloride group(YG),the Qiangxin capsule high-dose group(QG)were higher than the model group(MG),the difference was statistically significant(P<0.05);(4)Comparison of left ventricular mass index(LVMI):Compared with the normal group(KG),the index of the other groups were increased in different degree;The index of the model group(MG)and the Qiangxin capsule low-dose group(QD)were obviously higher than the blank group,the difference was statistically significant(p<0.01);The index of the normal group(KG),the trimetazidine hydrochloride group(YG),the Qiangxin capsule high-dose group(QG)were higher than the model group(MG),the difference was statistically significant(P<0.05 or P<0.01);(5)General observation of rats: In the course of intragastric administration,rats in the normal group(KG)had good mental state and had a sensitive response;Rats in the model group(MG)had poor sperm status,unresponsiveness,and mutual biting;The rest of the rats were in a state of mental state good and eating normally.Conclusion:Qiangxin Capsule can increase the activity of Ca2+-Mg2+-ATPase in myocardial cells and improve myocardial energy metabolism.Its mechanism of action in delaying heart failure may be related to improving myocardial energy metabolism.
