Objective: Research Team carry out this study in order to probe the effects and mechanisms of "Yunpi Powder",which is an empirical formula for infantile anorexia created by Professor Huang Jianye,a national distinguished veteran paediatrician of traditional Chinese medicine(TCM),to the gastrointestinal tract and learn the influence of Yunpi Powder on gastrointestinal motility and gastric mucosa of anorexia rats and its regulation function of the gastrointestinal hormones such as serum motilin and amylase.The research results will provide theoretical basis for the clinical treatment to infantile anorexia as well as popularization and application of empirical formula of distinguished veteran doctor of TCM.Methods: Choose 56 healthy 35 to 40-day-old Sprague Dawley rats,weighed(60±10)gram,with half male and half female,randomly divided 16 rats into normal group(group A)and 40 rats into model group after adaptive feed.Feed the model rats with specific fodder for 1 week,observe and record the change of appetites and weights of rats.After successful modeling,choose rats from normal group and model group,8 of each and draw materials.Divide the remaining 32 rats randomly into 4 group: Blank Group(Group B),Yunpi Powder Group(Group C),Xingpi Yanger Granule Group(group D),Normal Saline Control Group(Group E),with 8 in each group(half male and half female).each group feed whit normal fodder,Meanwhile,each group was given a gavage as treatment for 7 days.Group A and Group B was given the same volume of distilled water,Group C was given a gavage with Chinese medicine liquid of Yunpi Powder;Group D was given a gavage with Chinese medicine liquid of Xingpi Yanger Granule,Group E was given a gavage with the same volume of Normal Saline.Draw materials after 1-week treatment.Observe the effect on serum motilin,amylase,gastrointestinal and gastric mucosa of the model rats and treated rats.Results:(1)Compared with the normal rats Group A,the model rats have poor appetite and less weight(P<0.01),which prove successful model.(2)After treatment,amounts of food intake of rats grow in the short term,and quickly returned to normal rats group A for Yunpi powder C group followed by D group,B group,E group;after treatment,weights of food intake of rats grow in the short term,and quickly returned to normal rats group A as group D second for Yunpi powder C group,B group and E group.(3)Serum amylase and serum motilin: the Serum amylase and motilin of model rats decreased significantly(P<0.01);after treatment,the Serum amylase and motilin of rats in Group C increased significantly,Group D increased secondarily(P <0.01),Group B and Group E have no difference after treatment(P>0.05).(4)Gastrointestinal motion: Model group rats gastrointestinal propulsion function decreased significantly(P < 0.01);after treatment,the gastrointestinal motion of rats in Yunpi powder C and Xingpi Yanger Granule Group D is hypofunctional(P<0.01).Group B and Group E have no difference after treatment(P>0.05).(5)The change of the electron microscope observation of gastric mucosa in mitochondria,endoplasmic reticulum,nucleus,villus ultrastructure: The structure of cells from Group A is complete with a large number of microvilli distributions and the structure of mitochondria,endoplasmic reticula is clear.Gastric mucosa cells form model rats have the nuclei without nucleolus and large a amount of vesicular organization.Lipid droplets can be seen and Gastric microvilli cannot be seen.the Structure of Mitochondria and endoplasmic reticulum is oedematous and incomplete.After treatment,Yunpi powder C Group: the mitochondria,endoplasmic reticula and nucleoli quickly returned to normal rats group A.Group D: the mitochondrial cristae are not obvious.Some mitochondria have autolysis phenomenon and some nucleus have no nucleoli.Group B: the mitochondrial cristae are not clear.The gaps of Endoplasmic reticula are wide and endoplasmic reticula fracture.The ratio of nucleus and cytoplasm is abnormal.The nuclei have no nucleoli.Group E: The cells contains a small amount of lipid droplets.Part of intercellular substance Fibrosis.Nucleus gaps widened and have no nucleoli.Conclusion: 1.Feeding rats with specific fodder can make anorexia rats mode successfully.This method is feasible;2.The main reason of anorexia is the reduction of serum motilin and amylase and the hypofunction of gastrointestinal motion.Anorexia can the decompose of the electron microscope observation of gastric mucosa in mitochondria,endoplasmic reticulum,nucleus,villus ultrastructure;3.The decrease of gastrointestinal hormone secretion and the decrease of gastrointestinal impetus may be related to the ultrastructural damage of gastric antrum mucosa;4.Yunpi powder in the treatment of anorexia rats in the C group to promote serum motilin and amylase secretion,enhance the gastrointestinal impetus,repair of gastric mucosa ultrastructure of rats and food intake,weight recovery etc,were significantly better than the other treatment groups;5.The mechanism of the treatment of anorexia in the treatment of anorexia may be achieved by repairing the ultrastructure of gastric antrum mucosa,promoting the secretion of gastrointestinal hormones and enhancing the impetus of stomach and intestine.The Yunpi Powder for anorexia is worth popularization and application.
