Objective:To observing the effects of different dosage Huanglian Jiangtang Tablet on fasting blood glucose,fasting insulin,insulin resistance index,GLP-1,blood lipid,function of liver and kidney in type 2 diabetic rats,and investigate the dose effect relationship,effectiveness,safety and the possible mechanism,for clinical safety and effective application of this medicine.Methods:Male SD rats were randomly divided into model group and normal group,model group fed with high fat diet for 4 weeks,intraperitoneal injection of 45mg/kg1%STZ,to induce the model of type 2 diabetic rats,the normal group received routine feeding and intraperitoneal injection of the same volume of citric acid buffer.Diabeticratswererandomlydividedinto5groups,respectivelyfor huanglianjiangtang tablets in low dose group,middle dose group,high dose group(respectively 0.162,0.324,0.648 g·kg-1 of crude drug),positive control group(Metformin Hydrochloride Tablets 0.2g kg-1),and the model group.Even the huanglianjiangtang tablets different dosage group and positive group were given the specified dose with gavage treatment,model group and normal group given the same volume of distilled water.After 4 weeks,the blood glucose of tail vein was tested once a week.After 4 weeks of treatment,all rats were anesthetized with chloral hydrate,draw blood from the abdominal aorta,test FBG,FINS,GLP-1,blood lipid,liver function,and renal function,and calculate the HOMA-IR.Results:1.After injection molding,the weight of rats in the model group decreased(P<0.05),FBG was significantly higher than that in the normal group(P<0.05).2.After treatment for 4 weeks,the FBG levels in the rats of the three different dosage Huanglian Jiangtang Tablet groups were significantly decreased compared with the model group(P<0.05),and with the increase of the drug dose,the level of the blood glucose decreased significantly.3.After treatment for 4 weeks,FINS,HOMA-IR andGLP-1 in the model group were difference compared with the normal group(P<0.05);The FINS level in low and middle dose group of huanglianjiangtang tablets was not significantly higher than that in model group(P>0.05),high dose group FINS levels were significantly increased compared with the model group(P<0.05).The HOMA-IR of huanglianjiangtang tablets three dose group compared with model group decreased significantly(P<0.05),the GLP-1 level was significantly increased compared with the model group(P<0.05),with the increase of the drug dose,the level of the GLP-1decreased significantly.4.After treatment for 4 weeks,the lipid in model group were difference compared with the normal group(P<0.05),the HDL-C in three dose group of huanglianjiangtang tablets had no difference compared with the normal group(P>0.05),but the LDL-C,TG,and TC had significant difference compared with model group(P<0.05).5.After treatment for 4 weeks,the model group liver function and renal function compared with the normal group had significantly increased(P<0.05),liver function in three dose group of huanglianjiangtang tablets had no difference compared with the normal group(P>0.05).Conclusion:High fat diet for 4 weeks,according to the weight give 45mg/kg STZ to the rats by intraperitoneal injection,to successfully establish the rat model of type 2diabetes.The treatment of type 2 diabetic rats in different doses of huanglianjiangtang tablets can get a dose-effect relationship,and good safety.The mechanism about huanglianjiangtang tablets reduce blood glucose may be related to decreasing insulin resistance,increased insulin and GLP-1 secretion.
