Objectives: To investigate the association of plasma miR-1and ischemic stroke in patients with nonvalvular atrial fibrillation(AF).Methods: 1)Use case-control study,78 patients were selected in the First Affiliated Hospital of University of South China from April,2016 to December,2016.These patients were devided into two groups: 30 cases in experimental group(AF with ischemic stroke),and 48 cases in control group(AF without ischemic stroke).The conditions of sex,age,smoking,drinking and medical history in each group were well matched.Patients in experimental group have been underwent cranial CT or MRI examination to determine ischemic stroke diagnosis.2)The venous blood samples were collected and anticoagulated by EDTA in the next morning after admission.The serum sample was stored in-80℃ refrigerator.The expression level of plasma miR-1 was detected by quantitative polymerase chain reaction(qRT-PCR).The expression level of GPⅡb/Ⅲa and P-selectin was detected by enzyme-linked immunosorbent assay(ELISA).Left atrial diameter(LAD)was examined by echocardiography.3)We recorded and analyzed the general data,blood lipid levels(TC、TG、LDL-C、HDL-C),left atrial diameter(LAD),plasma miR-1,GPⅡb/ IIIa,and soluble P-selectin levels in experimental and control group.Therelationships between plasma miR-1 and GPⅡ b/ Ⅲ a,soluble P-selectin,age,blood lipid levels(TC 、 TG 、 LDL-C 、 HDL-C)and LAD in experimental group were analyzed.Results: 1)Sex,age,smoking,drinking,medical history,the level of blood lipid levels(TC、TG、LDL-C、HDL-C),and LAD aren’t statistically different between experimental and control group.2)Compared with control group,the expression level of plasma miR-1,GP Ⅱ b/ Ⅲ a,and soluble P-selectin in patients of experimental group is higher,and the difference is statistically significant.3)In experimental group,plasma miR-1 is positively correlated with GP Ⅱ b/ Ⅲ a and P-selectin.And GP Ⅱ b/ Ⅲ a is positively correlated with P-selectin.Conclusions : 1)Up-regulation of the plasma miR-1 level is associated with the occurrence of ischemic stroke in patients with nonvalvular atrial fibrillation,and it may be involved in platelet activation.2)GPⅡb/Ⅲa and P-selectin may be used as biomarkers for patients with nonvalvular atrial fibrillation complicated by ischemic stroke. |
