Immune Effect Of Serum Interleukin-10、interleukin-12and Interferon-γ In The Pathogenesis Of Brucellosis | Posted on:2018-12-13 | Degree:Master | Type:Thesis | Country:China | Candidate:H H Lv | Full Text:PDF | GTID:2334330536469688 | Subject:Internal Medicine | Abstract/Summary: | PDF Full Text Request | Objective By detecting the IL-10、IL-12 and IFN-γ level of the brucellosis patients before treatment and at the time of one cycle and three cycles after the treatment,we can research their connection with brucellosis attack,and discuss how they work in the immune function of the process of brucellosis.Methods(1)we choose 30 rucellosis patients who were not been treated in infectious department of Ningxia Medical University General Hospital as our research object,averaged(50.9±13.6),they are aged between 13-73 years old who were given 3 cycle’s formal treatment.All the diagnoses are all up to the diagnosis standard the eighth edition of the textbooks,Which are published by our country’s People Medical Publishing Company.Another 30 healthy persons were selected as control group.(2)Using double antibody sandwich enzyme-linked immunosorbent assay(ELISA)to detect serum IL-10,IL-12,IFN-γlevel of the patients with brucellosis and the healthy control group before treatment and 1cycle and 3 cycles after the treatment.At the same time,we follow-up and observed the clinical prognosis of patients with brucellosis,and regularly review brucella agglutination test.(3)What’s more,SPSS 22 statistical software was used to analyze the levels and changing feature of serum IL-10,IL-12,IFN-γ,and its relationship with the corresponding cytokines in the control group.Results 1.The serum IFN-γ level of patients with brucellosis before treatment is(26.47±37.65)pg/ml,which is obviously higher than the level of control group((3.15±0.51)pg/ml),the difference was statistically significant(P<0.01);Serum IFN-γ level after one cycle and three cycles’ treatment are respectively(3.10±0.52)pg/ml、(3.12±0.52)pg/ml,theyare not statistically significant(P >0.05)compared with the control group.There were statistically significant differences(P<0.01)when compared in different clinical stages and among multiple groups;after one and three cycles of treatment,the serum IFN-γ of the treatment group decreased significantly compared with before treatment,which was statistically significant(P <0.01).2.The serum IL-12 level of patients with brucellosis before treatment is(17.69±15.97)pg/ml,which is obviously higher than the level of control group((4.54±1.07)pg/ml),the difference was statistically significant(P<0.01);Serum IL-12 level after one cycle and three cycles’ treatment are respectively(4.49±0.83)pg/ml、(4.30±0.91)pg/ml,they are not statistically significant(P >0.05)compared with the control group.There were statistically significant differences(P<0.01)when compared in different clinical stages and among multiple groups;after one and three cycles of treatment,the serum IL-12 of the treatment group decreased significantly compared with before treatment,which was statistically significant(P <0.01).3.The serum IL-10 level of patients with brucellosis before treatment,after one cycle and three cycles’ treatment are respectively(7.62±6.15)pg/ml、(5.63±2.59)pg/ml、(4.46±1.54)pg/ml,which showed no significant difference compared with the control group(P>0.05).At the same time observed IL-10 levels of 30 cases of patients with brucellosis before treatment,of which 8 cases(26.67%)was significantly higher than the average level of health control group IL-10(7.62pg/ml).The subject has fellow-up and observated the clinical outcome of 30 patients with brucellosis further more,and we found brucella agglutination test of 5 cases(62.50%)from 8 cases elevated IL-10 levels is still positive and clinical symptoms were not relieve after three to five months of regular drug treatment.7 cases(31.82%)from the remaining 22 cases is still positive and clinical symptoms were not relieved after three to five months of regular drug treatment after the review of brucella agglutination test.4.The serum IFN-γ,IL-10 level of patients with brucellosis are respectively(26.47±37.65)pg/ml、(17.69±15.97)pg/ml,they showed a positive linear relationship(r=0.886,P<0.05).Conclusion 1.The expression of cytokines in patients with brucellosis showed a major immune response bias for Th1 type.2.IFN-γ and IL-12 may have some anti-inflammatory role in the pathogenesis of brucellosis,and the anti-inflammatory effect of IL-12 may be through the IFN-γ dependent pathway. | Keywords/Search Tags: | Brucellosis, Interleukin-10, Interleukin-12, Interferon-γ | PDF Full Text Request | Related items |
