Objective To study the polymorphisms of HLA-B and DRB1 in Hui and Han children from Ningxia,the distribution characteristics of HLA-B and DRB1 alleles in Hui and Han children from Ningxia were analyzed by calculation and analysis,and compared with the TCM constitution to explore the association between polymorphism of HLA and TCM constitution,in the same time,we compared with other populations of the relevant alleles,expect to provide an objective laboratory basis for the incidence and prognosis of children with Henoch-Schonlein purpura and purpuric nephritis in the region.Method Collection of blood samples from 76 children of Hui and Han from Ningxia,detection of HLA-B,DRB1 genotypes by PCR-sequence specific primers(PCR-SSP)method,statistical analysis of the results of classification,and the results were analyzed and compared with other populations.At the same time,the volunteers were divided of TCM constitution,the HLA data and TCM constitution for statistical analysis.Results 1.There were 19 alleles of HLA-B were detected in Han from Ningxia,Gene frequency from high to low were B*15(16.3%)、B*35、40(13.0%)、B*13、46(9.8%)、B*44、51(5.4%)、B*50(4.3%)、B*07、37、48、52(3.3%)、B*08、55(2.2%)、HLA-B*18、38、49、54、57(1.1%).There were 13 alleles of HLA-DRB1 were detected in Han from Ningxia,Gene frequency from high to low were DRB1*12(19.6%)、DRB1*09(16.3%)、DRB1*07(12.0%)、DRB1*04、14(9.8%)、DRB1*15(8.7%)、DRB1*08(7.6%)、DRB1*13(4.3%)、DRB1*11、16(3.3%)、DRB1*03、10(2.2%)、DRB1*01(1.1%).2.There were 19 alleles of HLA-B were detected in Hui from Ningxia,Gene frequency from high to low were B*40、46(18.3%)、B*15(11.7%)、B*13(8.3%)、B*07、35、44、48、51(5.0%)、B*58(3.3%)、B*08、37、38、39、45、49、52、54 、 55(1.7%).There were 12 alleles of HLA-DRB1 were detected in Hui from Ningxia,Gene frequency from high to low were DRB1*12(16.7%)、DRB1*15(15.0%)、DRB1*04、14(13.3%)、DRB1*11(11.7%)、DRB1*07、09、13(6.7%)、DRB1*03、08(3.3%)、DRB1*01、16(1.7%).3.HLA-B and HLA-DRB1 allele frequency distribution in Han and Hui from Ningxia populations were similar,the difference was not statistically significant.4.The HLA-B and DRB1 alleles of Han from Ningxia population are similar to those of Han from Shanxi and Han from Shandong Han population,but they also have their own distribution characteristics.Conclusions 1.HLA-B and HLA-DRB1 allele frequency distribution in Han and Hui from Ningxia populations were similar,the difference was not statistically significant.2.The HLA-B and DRB1 alleles of Han from Ningxia population are similar to those of Han from Shanxi and Han from Shandong Han population,but they also have their own distribution characteristics.3.It is speculated that HLA-DRB1*01/11 is a susceptible gene for children with HSP/HSPN in Han from Ningxia,HLA-DRB1*07 is a protective gene;HLA-B*35 、 HLA-DRB1*01 is a susceptible gene for children with HSP / HSPN in Hui from Ningxia,HLA-DRB1*07is a protective gene.4.HLA has obvious ethnic and regional differences,which provides important reference for population genetics and HLA disease related research.
