| Objective To study the Impact of Hydrogen Sulfide on ERK,JNK and p38 m RNA expression level of MAPK signal transduction pathway of skin macrophages of burned rats cultured in vitro and to demonstrate the molecule mechanism of H2 S in wound healing.Method Skin macrophages from burned rats were the research object.To select 5 rats from 30 healthy SD rats and receive no intervention and the other 25 rats were formed a model of 5%TBSA deep second degree burns(confirmed by biopsy).Randomly choose a rat from 5 normal rats as blank control group and to select 12 rats from 25 burn rats and randomly divide into 4 groups: burn control group,GLBN group,Na HS group and Na HS+GLBN group(3 rats in each group).After creating burn models separate macrophages from the normal back skin and burned wound into culture flasks.After culturing to get stable,appropriate drugs were added respectively into culture flasks and the final concentration of the drugs were 200umol/L(add equivalent PBS solution into blank control group and burn control group as controls).After adding the drug the change process and its significance of ERK,JNK and p38 m RNA in the macrophages were determined with RT-PCR at these three time points of 1h,6h,12 h.Results To compare the expression result of ERK m RNA in each group: there was statistically significant difference between black control group and burn group(P<0.05);compared with other groups,the Na HS group had statistically significant difference at the same point(P<0.05);at the same point,compared Na HS+GLBN group with GLBN group,there was statistically significant difference about the expression of ERK m RNA(P < 0.05);there was significant difference in the expression of ERK m RNA between GLBN group and burn control group(P < 0.05).To compare the expression result of JNK m RNA in each group: there was statistically significant difference between black control group and burn group (P<0.05);compared with other groups,the Na HS group had statistically significant difference at the same point(P<0.05);at the same point,compared Na HS+GLBN group with GLBN group,there was significant difference about the expression of JNK m RNA(P < 0.05);there was significant difference in the expression of JNK m RNA between GLBN group and burn control group(P < 0.05).To compare the expression result of p38 m RNA in each group: there was statistically significant difference between black control group and burn group(P<0.05);compared with other groups,the Na HS group had statistically significant difference at the same point(P<0.05);at the same point,compared Na HS+GLBN group with GLBN group,there was statistically significant difference about the expression of p38 m RNA(P < 0.05);there was significant difference in the expression of p38 m RNA between GLBN group and burn control group(P < 0.05).Conclusion Hydrogen sulfide increases the relative expression level of ERK m RNA and decreases the expression level of JNK and p38 m RNA to affect the transmission of MAPK signal pathway of the burn rat skin macrophage cultured in Vitro by regulating and controlling the K+ channel. |
