The Effect Of CO2 Pneumoperitoneum On Pancreas,IL-1,IL-6 And Ia Antigen In Rats With Severe Acute Pancreatitis In Xining Area | | Posted on:2018-09-25 | Degree:Master | Type:Thesis | | Country:China | Candidate:K F Su | Full Text:PDF | | GTID:2334330536464151 | Subject:Surgery (General Surgery) | | Abstract/Summary: | PDF Full Text Request | | Objective:To investigate the effect of laparoscopic CO2 pneumoperitoneum on pancreatic tissue,blood amylase,expression of IL-1,IL-6 and Ia antigen in rats with severe acute pancreatitis,and to provide the experimental basis for the application of laparoscopy in the treatment of SAP.Methods:36 healthy male SD rats were randomly divided into 3 groups by random number table method: 1.Sham-operation group(n=12): Only open the abdomen and gently flip the duodenum and pancreas,the abdomen was closed after 30 minutes.2.SAP group(n=12): Multiple point injection of subcapsular pancreas with 5% taurocholic acid sodium salt was used to prepare SAP rat model,and then abdomen was closed without charging CO2.3.SAP+CO2 pneumoperitoneum group(n=12): The same method was used to prepare SAP rat model and close the abdomen.6 hours later,veress needle is placed by abdominal puncture,CO2 was injected into abdominal cavity of rats for 2 hours,with a pressure of 8mm Hg.The animals were sacrificed 8 hours after operation in each group.The abdomen was opened for the blood from inferior vena cava.The serum amylase was detected by automatic biochemical analyzer.Enzyme linked immunosorbent assay(EILSA)and flow cytometry were used to detect the expression of serum IL-1,IL-6 and Ia antigen in peripheral blood B cells of rats.The pancreatic tissues were cut down and performed pathological examination by ordinary light microscopy.The data of three groups were compared and analyzed statistically.Results: 1.Compared with the sham-operation group,the content of serum amylase in SAP group and SAP+CO2 pneumoperitoneum group was significantly higher.The difference was statistically significant,P<0.05.Compared with the SAP group,there was no significant difference in the content of serum amylase in SAP+CO2 pneumoperitoneum group.The difference was not statistically significant,P>0.05.2.Compared with the sham-operation group,the level of serum IL-1,IL-6 in SAP group and SAP+CO2 pneumoperitoneum group was significantly higher.Thedifference was statistically significant,P<0.05.Compared with the SAP group,the level of serum IL-1,IL-6 in SAP+CO2 pneumoperitoneum group was not significant.The difference was not statistically significant,P>0.05.3.Compared with the sham-operation group,the expression of Ia antigen in peripheral blood B cells of rats was significantly decreased in SAP group and SAP+CO2 pneumoperitoneum group.The difference was statistically significant,P<0.05.Compared with the SAP group,the expression of Ia antigen in peripheral blood B cells of rats in SAP+CO2 pneumoperitoneum group was not significant.The difference was not statistically significant,P>0.05.4.Compared with sham operation group,HE staining of pancreatic pathology of SAP group and SAP+CO2 pneumoperitoneum group show Pancreatic necrosis,inflammatory cell infiltration and hemorrhage.Pathological score was statistically significant,P<0.05.Compared with SAP group,Pathological score of SAP+CO2 pneumoperitoneum group was not statistically significant,P>0.05.Conclusion: In Xining area 1.The establishment of CO2 pneumoperitoneum has not obvious influence on pancreatic pathology and serum amylase of severe acute pancreatitis rats.2.The level of IL-1 and IL-6 of SAP rats has not been obvious influenced by the influence of CO2 pneumoperitoneum.3.SAP group showed significant immunosuppression,The establishment of CO2 pneumoperitoneum has not obvious promoted increased Ia antigen suppression。... | | Keywords/Search Tags: | CO2 pneumoperitoneum, severe acute pancreatitis,SAP, Interleukin 1,6, Ia antigen | PDF Full Text Request | Related items |
