Objective To study Tibetan snow lotus plant polysaccharide could protect human immortalized cutin forming cells(Ha Ca T cells)which was cultured in vitro from oxidative damage coused by Ultraviolet radation b(UVB)induction.Method Apply the method of the invitro cells culture experimental to culture the immortalized human epidermal cell(Ha Ca T cells),according to the different of radiation energy,divided the cells into high radiation group and low radiation group.Application different concentrations of Tibet snow lotus polysaccharide which was separated by people to intervene on the radiation cells.The cells were divided into control group(normal training,no radiation,no polysaccharides),the radiation group(radiation,no polysaccharides,low concentration polysaccharide group and high concentration of polysaccharide group.Polysaccharide1 was added into cells culture ahead of time 1h.The cells of each group need to culture 18 h after irradiation.Applying trypan blue staining method for each group to observe the morphology and apoptosis after giving radiation condition and taking photographs.The MTS method was applied to measure cell proliferation;Determination of antioxidant enzymes: GSH(GSH)and associates of MDA(malondialdehyde)content,visible spectrometry CAT(catalase)and the WST-1 method determining SOD(superoxide dismutase(SOD)activity.Result Compared with radiation groups,giving the different concentrations of polysaccharide,the number of cells who was disseminated by trypan blue were descended,integrity of cellular structure was obvious recovery,the mortality of cells was reduced;Proliferation activity was increased;the content of MDA and GSH was reduced,The activity of SOD and CAT was increased.Conclusion After radiated by different energy of UVB,giving a certin dose of polysaccharide to Ha Ca T cells could reduce the cellular structure damage and apoptosis caused by light damage.
