Objective: To study the inhibitive effects and mechanism of Polysaccharide of Semen Plantaginis on the free radical and nonenzymatic glycosylation in aging model rats induced by D-galactose.Methods: Fifty Wistar rats were randomly divided into 5 groups , the youth compares group , the senile model group and three model medication administration groups(the polysaccharide large dose group ; the polysaccharide small dose group and the apozema group ) . Senile model group and three model medication administration groups were daily nucha hypodermically injected D-galactose for 42 days to induce aging model .Then the youth compares group and the senile model group were drenched warm boiled water, and three model medication administration groups were respectively given polysaccharide of Semen Plantaginis according to 0.8g/kg.bw, 0.3g/kg.bw and apozema of Semen Plantaginis according to 15g/kg.bw for 40 days.Simultaneously killed rats , measured the capability of T-AOC and inhibit hydroxy radical, the activity of MAO-B , GSH/GSSH , carbonylation protein level in cerebral cortex . And the expression of RAGE-mRNA and AR-mRNA was determind by RT-PCR .Results: Compared with the young group , the senile model group both the carbonylation protein level , the activity of MAO-B , the expression of RAGE-mRNA and AR-mRNA were increased (P<0.01). But the capability of T-AOC , inhibit hydroxy radical and GSH/GSSH were decreased significantly (P<0.01) . Compared with the senile model group , the capability of T-AOC , inhibit hydroxy radical and GSH/GSSH were increased significantly(P<0.05,P<0.01) in three model medication administration groups , while the carbonylation protein level , the activity of MAO-B , the expression of RAGE-mRNA and AR-mRNA were decreased (P<0.05,P<0.01). The effects of the polysaccharide large dose group is best obvious than other medication administration groups .Conclusion: These results suggest that Semen Plantaginis have anti-aging contribution by elevating the body antioxygen capability and carbonyl group cleaning capability . Polysaccharide of Semen Plantaginis is the main significant ingredient .
