Objective: According to the different grades of glioma tissues and normal brain tissues Smurf2 protein expression were determined to investigate its expression in human gliomas and normal brain tissues and the significance of the related mechanism.Methods: All patients with glioma specimens were taken from September 2016 to January 2016 in the second hospital of Hebei Medical University,Department of Neurosurgery,surgery,30 cases were classified by the hospital pathology kegaonian endowment physicians according to classification standard of WHO authority,the WHO I level of 8 cases of glioma,10 cases of glioma WHO grade II,WHO grade III gliomas in 7 cases,5 cases of WHO grade IV gliomas.The glioma specimens collected in this study are the first diagnosis and have not been treated with radiotherapy,chemotherapy and related tumors.At the same time,6 normal brain tissue samples of patients with traumatic brain injury were collected and served as control group.All the collected specimens were stored in 10% formalin and liquid nitrogen tanks in 10 minutes after two minutes in vitro,and the errors in the sampling process were reduced to the greatest extent.According to the level of glioma and control group were randomly divided into five groups,using IHC(immunohistochemistry)method for detection of expression of Smurf2 in different pathological specimens,collected data,and the expression of Smurf2 in each sample were collected and analyzed by statistical software SPSS13.0.The Western Blot experimental method of Smurf2 in each sample were measured and Western blot,using Quantity One software test strip gray value,finally carries on the statistics and in the use of SPSS 13 software analysis.Result:1.Immunohistochemical test results: statistical analysis consistent with the normal distribution: Smurf2 in normal brain tissue and(I ~ IV)glioma group at all levels of the positive rate of the results are as follows: normal brain tissue group(n = 6): 11.96 ± 9.38%,WHO grade I glioma group(n = 8):28.23 ± 10.01%,WHO II glioma group(n = 10): 45.05 ± 12.53%,WHOIII grade glioma group(n = 7): 65.50 ± 16.17%,WHOIV grade glioma group(n= 5): 84.82 ± 13.33%.The expression of Smurf2 was higher than that of the control group,and the correlation coefficient was r = 0.894,P <0.01,and the expression of Smurf2 was correlated with the tumor grade according to the statistical analysis of Spearman rank correlation analysis.2 Western blot test results: The results were as follows: normal brain tissue group(n = 6): 0.1307 ± 0.07124,WHOI glioma group(n = 8): 0.2287 ±0.05592,WHOII glioma group(n = 10): 0.4120 ± 0.06812,WHOIII glioma group(n = 7): 0.6014 ± 0.06914,WHOIV grade glioma group(n = 5): 0.8640± 0.07635,the glioma group at all levels and the control group were compared,P <0.05,all of which were statistically significant.Conclusions:1 Compared with normal brain tissue,the expression of Smurf2 in all grades of gliomas was significantly up-regulated.(P <0.01).2 The higher the degree of malignant tumor,the higher the content of Smurf2 protein,and the expression of protein is closely related to the tumor grade,suggesting that the malignant degree of glioma may affect the protein expression of Smurf2 to a certain extent.
