Objective: Gastric cancer is a common gastrointestinal malignancy, with the characteristics of high morbidity and high mortality [1].China is a country of high incidence of gastric cancer, the pathological type of which is mainly adenocarcinoma-based and occasionally squamous cell carcinoma, squamous cell carcinoma and carcinoid [2].To these diseases, surgical treatment is the best for early stage, but because of the lack of specificty in the early symptoms, patients with gastric cancer couldn’t be discovered in the early period,so 60%-70% of patients have lost the chance of surgery when they were diagnosed. For the advanced patients, still more than half of patients who would suffer from recurrence and metastasis in five years after surgery,so chemotherapy and targeted drug therapy and other comprehensive treatment are the current treatments and develop certain effect. At present, the commonly used chemotherapy drugs for gastric cancer include paclitaxel, docetaxel, irinotecan, oxaliplatin, cisplatin and fluorouracil. We found that the sensitivity of different gastric cancer patients to paclitaxel combined with S-1 chemotherapy regimens are different. There are study found that the expression levelis of β-tubulin-Ⅲ is relative with the sensitivity of the chemotherapy of paclitaxel-related drugs. The positive expression of BRCA1 in high degree is one of the reason for the platinum resistance,It suggests that the expression of β-tubulin-Ⅲ and BRCA1 has relationship with the resistance of paclitaxel-related drugs.research on β-tubulin-Ⅲ and BRCA1 in breast cancer and ovarian cancer are common but not much ingastric cancer. It suggests that the expression of β-tubulin-Ⅲ and BRCA1 has relationship with the resistance of paclitaxel-related drugs. The aims of this study are to investigate the relationship between the expression of β-tubulin-Ⅲ and BRCA1 in gastric carcinoma and the clinicopathological features of gastric cancer and the relationship between the expression of β-tubulin-Ⅲ and BRCA1 and the chemotherapy of paclitaxel combined with S-1 chemotherapy regimens and to tentatively discuss their value in the development an prognosis of gastric cancer to achieve individual treatment.Method: This is a retrospective study by using Immunohistochemical SP method to test β-tubulin-Ⅲ、BRCA1 in 50 cases. The 50 samples of the gastroscopy are all from Harrison International Peace Hospital from January,2010 to January 2012.The pathological pattern is mainly adenocarcinoma.According to the degree of tumor differentiation,24 cases are low differentiation and 26 cases are high differenti-ation,Among these patients, there are 41 males and 9 females.in 50 patients.Their ages range from 32 to 78,and the median is 58 years old.The number of patients who are older than 60 is 22,and the others are younger. These patients all receive chemotherapy with Taxanes and S-1 for 2-8 periods(median is 4 periods),and the follow-up records are complete. The aims of this study are to investigate the relationship between the expression of β-tubulin-Ⅲ and BRCA1 in gastric carcinoma and the clinicopathological features of gastric cancer and the relationship between the expression of β-tubulin-Ⅲ and BRCA1 and the chemotherapy of paclitaxel combined with S-1 chemotherapy regimens and to tentatively discuss their value in the development an prognosis of gastric cancer to achieve individual treatment..Results: There are 37 cases with negative expression of β-tubulin-Ⅲ and the others with positive expression in 50 patients. As for BRCAI,the result is negative in 32 cases and positive in 18 cases. The partial response(PR)was 17,the stable disease(SD)rate was 15,and the progress disease(PD)was 18.The expression of β-tubulin-Ⅲ and BRCA1 on the patients who suffer from gastric cancer is not related to the age and gender of the patient.Statistical analysis shows that The positive expression of BRCA1 in low degree of differentiation was significantly higher than that in high degree of differentiation. The response to taxanes in β-tubulin-Ⅲ negative expression group was significantly higher than that in positive group(P=0.002).Conclusion: The expression of β-tubulin-Ⅲ and BRCA1 on the patients who suffer from gastric cancer is not related to the age and gender of the patient. The positive expression of BRCA1 is related to degree of differentiation. The positive expression of β-tubulin-Ⅲ is related to the sensitivity of the chemotherapy of paclitaxel and S-1.
