Objective: To comfirm weather salidroside(Sal)plays a part in protecting myocardial cell through reducing the myocardial ischemia and the apoptosis pathway of both death receptors and mitochondria in acute exhausted rats.Method:Male SD rats randomly divided into 4 groups(n=6): control group(Con),exhaustive exercise group(EE),low-dose and high-dose Sal pretreatment exhaustived swimming group(SEL,SEH).Groups were given Sal solution(15 or 30mg/kg/d)or the amount of normal saline through intraperitoneal injection 15 days.Con,EE group 0.9%NaCl were given(3ml/kg/d)intraperitoneal injection for 15 days.After the Sal pretreatment,the Con group did not do any exercise.EE,SEL and SEH groups’ rats were forced to swim until they were exhausted,refer to the Thomas exhaustion criteria.The changes of ischemia modified albumin(IMA)in serum were observed by ELISA.TUNEL and semi quantitative analysis method was used to detect the myocardial cell apoptosis index(AI)and immunohistochemical method to detect myocardial Bax/Bcl-2.Western blot method was used to detect the expression of myocardial Fas Cyto-c,Caspase-3,Caspase-8,and Caspase-9.Result:1 In terms of the serum levels of IMA comparison.The EE group(41.24±4.848U/ml)and the SEL group(32.09±2.068U/ml)were both significantly increased than Con group(15.31±1.98U/ml,P<0.05),the SEH(18.85±2.178U/ml)and SEL group were reduced than the EE group(P<0.05),and the SEH group was significantly lower than that the SEL group(P<0.05).2 Apoptosis index(AI): The EE group compared with the Con group increased significantly(36.00±2.00 vs 27.00±4.32,P<0.05)%.Although the SEL group(35.60±8.26)% was lower than the EE group,it was notstatistically significant(P>0.05).The SEH group(23.83±3.812)% was the lowest than the EE and SEL group,the difference was statistically obviously(P<0.05).3 Immunohistochemical method to detect myocardial Bax.Compared with the Con group(42.83±10.03)%,EE group(66.60±6.73)%,SEL group(57.00±7.70)% Bax immunohistochemistry score were increased(P<0.05).The SEH group(44.60±10.26))% was reduced evidently than the EE and SEL groups(P<0.05),and not significant different from the Con group.4 Immunohistochemical method to detect myocardial Bcl-2.Compared with the Con group(52.25±14.22)%,the level of Bcl-2 expression of the EE(27.67±5.03)% and SEL groups(33.25±4.03)% was reduced significantly(P<0.05).The SEH group(56.00±13.51)% was increased obviously than the EE and SEL groups(P<0.05),and was slightly higher than the Con group but no statistical difference.5 The Fas,Cyto-C and Caspase-3,Caspase-8,Caspase-9 myocardial expression quantity of the EE group(0.554±0.082,0.865±0.190,0.731±0.107,0.711±0.125,0.488±0.114),the SEL group(0.296±0.021,0.528±0.121,0.497±0.072,0.517±0.111,0.291±0.134)and the SEH group(0.181±0.027,0.270±0.050,0.262±0.044,0.268±0.054,0.164±0.067)wre increased significantly than the Con group(0.102±0.032,0.163±0.019,0.143±0.033,0.153±0.033,0.091±0.032,P<0.05).The SEL group and the SEH group were reduced than the EE group(P<0.05).The Fas,Cyto-C and Caspase-3,Caspase-8 of the SEH group was reduced significantly than the SEL,and the difference between each other was statistically obviously(P<0.05).Conclusion: Salidroside play a role in protecting myocardial cell by reducing the myocardial ischemia and reducing myocardial cell apoptosis in exhaustive exercise rats.The mechanism of reducing myocardial cell apoptosis may be related to the decreased of Fas,Cyto-C,Caspase-3,Caspase-8,Caspase-9 and the increased of the Bcl-2. |
