Objective:In the great strength of military training, especially the new army soldier, there was a greater incidence of myocardial injury caused by exhausted exercise;The main performances were sudden cardiac death,severe cardiac arrhythmia, cardiac function reduce and abnormal myocardial enzymes, etc.It had the serious influence on the health of the army commanders;Therefore, it is necessary to research intensively heart damage caused by exhausted training. The problem that which have been studied and solved by many experts and scholars is How to reduce heart damage exhausted exercise and improve the compensatory function of the heart.Salidroside(Sal) which is the main medicinal components of rhodiola rosea.The effects Salidroside on the cardiovascular system mainly includes:enhance myocardial contraction force, against ischemia reperfusion myocardial injury,against hydrogen peroxide damage in myocardial cells, etc.Epinephrine and norepinephrine belong to catecholamine. Exhaustion exercise is a kind of intense stimulation; Myocardial produce excessive stress reaction; The sympathetic nervous was excited; And then adrenaline increased; It makes the coronary artery expansion, enhance myocardial contraction force, heart rate, myocardial damage at last. Studies have demonstrated that a moderate amount of norepinephrine on coronary artery diastolic blood vessels, and a high level of NE is harmful to the heart, can lead to coronary spasm, may play a leading role in the occurrence of myocardial ischemia.Ventricular pressure volume tube(P-V) is the commonly used method which can evaluate hemodynamic in recent years. The method is a quantitative analysis of cardiac function by left ventricular work and energy;And it has been widely used in various laboratory studies and clinical research.There was no report about the effect of Salidroside on the changes of hemodynamic in acute exhausted exercise rats by millar system.Study both at home and abroad had shown that heart rate variability (HRV) is reflection of cardiac autonomic nervous regulation.It is a simple and sensitive detection methods. Through analyzing R-R phase changes of sinus rhythm, it reflects the effect of neural and humoral factors on adjusting the sinoatrial node;And it ialso reflects the sympathetic nerve and the vagus nerve activity and the relationship between them. Currently there was no report about the effect of Salidroside on the changes of heart rate variability in acute exhausted exercise rats. Through observing heart rate variability in acute exhausted exercise rats after Salidroside intervention, the study is to explore the effect of Salidroside on protecting the heart.Therefore, this study is to evaluate heart damage in acute exhausted exercise rats and to explore the effect of Salidroside on protecting the heart. It observed the changes about myocardial markers,ECG, HRV and hemodynamic in acute exhausted exercise rats. To explore the effect of Salidroside on heart damage in acute exhausted swimming rats by adjusting the epinephrine and norepinephrine levels; To discuss application of traditional Chinese medicine on preventing cardiac injury; The study provided reference for prevention and treatment of cardiovascular disease and theoretical basis for further improving the army battle effectiveness.Methods:24 healthy male Sprague wrote-Dawley rats, randomly divided into 4 groups (n=6):the control group (Con), acute exhaustive swimming group (EE), Low dose salidroside-acute exhaustive swimming group (SEL), and hight dose salidroside-acute exhaustive swimming group (SEH). Groups were given Sal solution (15mg·kg-1·d-1,30mg·kg-1·d-1) or the amount of normal saline lavage 14 days. And the rats adaptability swam 3 times in pool, (20min/times), were drid fur; Then adaptively to record ECG of the rats. In formal experiment, the rats of the control group don’t swim.The rats of EE, SEL,SEH swam after feeding Salidroside 14 days.The rats were fast 12 hours before swimming. Keep the water temperature in 33℃, floating up and down not more than 1 ℃. The rats of EE group, SEL and SEH group were submitted to one-time exhaustive swimming with 3% body weight loads of tin wire in the tail, and stopped the exhaustive exercise when meeting the Thomas standard of exhaustion, and dried the fur. After exhaustive swimming the rats were anaesthetized at once.ELISA method for detection of serum brain natriuretic peptide (BNP), troponin T (TnT), troponin I (TnI), epinephrine (E) and norepinephrine (NE). Application of pressure volume catheter (millar catheter) to detecte hemodynamic change; Record limb II lead ECG of all group rats awaken and calculate the heart rate variability.Results:1 Compared with Con group, the serum TnT,TnI, BNP, E, NE of EE group were obvious rised, there are significant differences(P<0.05); Compared with EE group, the indexes of SEL group are lower, there were significant differences (P<0.05);2 Compare with the control group,the stroke volume(SV),left ventri cular end diastolic volume (Ved), time of isovolumic diastolic phase(Tau) in EE group increased (P<0.05); Left ventricular end systolic pressure(P es)in the EE group decreased (P<0.05).Compared with EE group,SV,Ved, Tau, Pes in SE groups had opposite changes (P<0.05)3 Compared with Con group,HR of EE group was accelerated, Q Amplitude was deepen,ST segment was raised,there were significant diff erences(P<0.01). Compared with EE group, R-R intervals,HR, ST segme nt of SEL group fell back, and all have significant difference (P<0.01).4Compared with Con group, TP, LF, HF, LF/HF of EE group were significantly reduced;The indexes of Sal group were increased significantly (P <0.05).Conclusion:Salidroside can effectively reduce the myocardial injury caused by acute exhaustive exercise, improve myocardial ischemia, improve the diastolic function of heart, and combat decreases of heart rate variability; Salidroside can protect myocardial injury probably by regulating the serum levels of epinephrine and norepinephrine. |
