| Objective: To study the effects of Chaihushugansan on the gastric recap- tor mRNA (GASR mRNA) and CCK-A receptor mRNA (CCK-AR mRNA) expression of stress rats induced by constantly squeezing their tails, we could to provide experimental evidence by revealing the method of soothing the liver and regulating qi treated gastrointestinal psychosomatic diseases and multi-target treating as a whole from the brain-gut axis and peptides angle.Methods: 30 rats were selected and randomly divided into 3 groups: control group, model group and Chinese herb group. (each n=10). The animal model of stressed was established by constantly squeezing their tails except normal group for 4wk, twice everyday , each time 30 min. From the first day , Chaihushugansan decoction contained 1.41g/ml drug and saline were administered respectively to Chaihushugan san group and normal group and model group by gavage (1.5ml/100g every time), twice everyday .The animal's general situation and amount of food and water were observed daily, body weight were measured before and after the experiment. In the 29th day, brain and Gastric tissue were taken for observing. Histological investigation of brain and Gastric tissue by HE staining. The expression of gastric receptor mRNA (GASR mRNA) and CCK-A receptor mRNA (CCK-AR mRNA) of brain and gastric tissue by In situ hybridization technique were detected. Results①the model group Model group appeared margination, pad batch, decline in excitement, activity and fight, getting sluggish, blunted in responsive- ness, loose and withered hair. The above manifestation in Chaihushugansan decoction were not so obvious compared with model group.②The amount of food and water in model group were less than those of normal group (both P<0.05), while the amount of food and water in Chaihushugansan group were more than those of model group (both P<0.05). Weight growth in model group were slower than those of normal group (P<0.05), weight growth in Chaihushu- gansan group were higher than those of model group, but there was no obvious difference.③Model group appeared slight to moderate stomach mucosa erosion, inflammation and hyperfunction of mucus secretion, but they were not so obvi- ous in Chaihushugansan group.④the GASR mRNA expression of the model group rats on the brain was increased significantly than the control group (P <0.05), while Chaihushugansan group decreased significantly than the model group(P<0.05). While CCK-AR mRNA didn't express in brain tissue.⑤GASR mRNA and CCK-AR mRNA expression of Model group were increased signify- cantly than the control group in gastric tissue (P <0.05), while Chaihushugansan group decreased significantly than the model group(P<0.05).Conclusions:①The method of soothing the liver and regulating qi has certain both physical and mental effect to be against the psychological stress.②Chaihushugansan decrease depression and dyspepsia,its mechanism is related to decreased expression of GASR mRNA on the brain and gastric tissues and CCK-AR mRNA on the gastric of rat. |
