| Objective:To investigate the expression of FLOT1 in gastric cancer samples and adjacent noncancerous tissue samples,even more,to analyze their correlation with pathological characteristics and prognosis.Methods:1.We detected the FLOT1 gene and protein expression in matched pairs of fresh gastric cancer samples and adjacent noncancerous tissue samples from the same patient by Real-time florescence quantitative PCR(qRT-PCR)(n = 16)and Western blot analysis(n=16).2.IHC was performed to examine FLOT1 protein expression in 157 gastric cancer paraffin-embedded tissues and 30 matched normal specimens from gastric cancer patients.Statistical analyses were applied to evaluate the associations of FLOT1 protein expression with clinical parameters and prognosis.Results:1、The expression of FLOT1 gene and protein in gastric cancer tissue(1)Compared with adjacent non-cancerous tissue,FLOT1 mRNA expression in gastric cancer tissue was significantly upregulated(3.12± 0.10 vs.0.81±0.11,P< 0.05)by Real-time florescence quantitative PCR.(2)Compared with adjacent non-cancerous tissue,FLOT1 protein expression in gastric cancer tissue was significantly upregulated(0.82±0.15 vs.0.23±0.11,P< 0.05)by Western blot.2、The relationship between FLOT1 protein expression and clinicopathologic parameters and prognosis of gastric cancer patients(1)The rate of elevated FLOT1 protein expression in gastric carcinoma tissue 68.15%(107/157)was significantly higher compared with that in adjacent noncancerous tissue 26.67%(8/30)by IHC(P< 0.05).(2)Expression of FLOT1 in tumor tissue was not significantly associated with gender(P=0.734),age(P=0.605),tumor size(P=0.391),differentiation status(P=0.152)and tumor location(P=0.531).However,elevated FLOT1 expression was strongly correlated with the depth of invasion(P = 0.000),lymph node metastasis(P = 0.000),distant metastasis(P= 0.000)and AJCC stage(P = 0.016).(3)The 5-year overall survival rate and disease-free survival rate in the high expression group was significantly lower than that in the low expression group(25% vs 57%,P<0.001;18% vs 51%,P <0.001).The difference was statistically significant between curves of FLOT1 high-expressing and low-expressing patients within subgroups of depth of invasion T3+ T4,lymph node metastasis N1-3 and AJCC stage III+IV.However,the difference was no statistical significance between curves of FLOT1 high-expressing and low-expressing patients within subgroups of depth of invasion T1+ T2,lymph node metastasis N0 and AJCC stage I+II.The multivariate Cox regression model showed that FLOT1 expression was a poor independent prognostic factor for survival time in gastric cancer patients.Conclusion:1、FLOT1 mRNA and FLOT1 protein levels was evidently up-regulated in gastric cancer tissues compared with that in adjacent non-cancerous tissue samples.Our date revealed the essential roles of FLOT1 in tumourigenesis of gastric cancer.2、Expression of FLOT1 in gastric cancer tissues was not significantly associated with gender,age,tumor size,differentiation status and tumor location.However,elevated FLOT1 expression was strongly correlated with the depth of invasion,lymph node metastasis,distant metastasis and AJCC stage.Our date revealed the essential roles of FLOT1 in invasiveness and metastasis of gastric cancer.3、Elevated expression of FLOT1 is an independent prognostic factor for GC patients. |
