| Objective:To investigate the clinical significance of mi RNA-338-3p in hepatocellular carcinoma tissue samples,and study the effects of mi RN A-338-3p expression on proliferation,migration,invasion,epithelial-to-mesenchymal transition of hepatocarcinoma cellsMethods:The expression of mi RN A-338-3p was evaluated on 35 pairs of HCC tissue samples and adjacent non-tumorous tissue by real-time PCR,then we analysed the significance of gene by comparing clinical data of patients.HCC cells(HCCLM3 and MHCC97-H)infected by lentivirus with mi RN A-338-3p gene,which could upregulating expression of gene in vitro.And we tested the effects of overexpression of mi RN A-338-3p on proliferation,migration,invasion and epithelial-to-mesenchymal transition of HCC cells by MTT assay,wound healing assay,transwell assay and Western Bolt.Results:Mi RN A-338-3p is downregulated in HCC tissue,which is related to differentiation degree,thrombus,tumor diameter,vascular invasion and postoperative survival(p<0.05).The MTT assay results are shown: absorbance of the experimental group(HCCLM3:1.153±0.232;MHCC97-H:1.247±0.247)are lower than the control group(HCC LM3:1.783±0.189;MHCC97-H:1.907±0.218)for 72 h.The wound healing assay results are shown: cells migration distance of the experimental group[HCCLM3:(89.7±30.7)μm;MHCC97-H:(164.7±33.5)μm]are lower than the control group(HCCLM3:(189.0±31.0)μm;MHCC97-H:(267.0±28.6)μm).The transwell assay results are shown: the number of invaded HCC cells of the experimental group(HCCLM3: 35.3±12.2;MHCC97-H: 74.7±10.6)are lower than the control group(HCCLM3: 70.0±17.1;MHCC97-H: 120.7±22.6).Western Bolt:It is not statistical different between E-cad and Vim protein expression level of the normal group and the control group.E-cad protein expression level of experimental group is higher than others,but Vim protein is lower.Conclusion: Mi RN A-338-3p is downregulated in HCC tissue by compared to adjacent non-tumorous tissue,and the patients with low level of this gene expression likely have poorer tumor prognosis and shorter survival time.In vitro,mi RN A-338-3p could suppress proliferation of HCC cells,and it may supress migration,invasion by inhibiting epithelial-to-mesenchymal transition in HCC cells. |
