| Obj ective:Substantial data indicate that oxidative stress is involved in the development of type 2 diabetes mellitus and diabetic retinopathy(DR).To investigate the distribution of genetic polymorphisms of biotransformation enzyme-glutathione S-transferase(GST)such as GSTM1 and GSTT1 polymorphism(313 A/G)alone and combined genotypes in type 2 diabetic patients and type 2 diabetic retinopathy patients by PCR,and to explore the relationship between these polymorphic genotypes and susceptibility of type 2 diabetic and type 2 diabetic retinopathy.Methods:In this study,selected the patients with type 2 diabetes mellitus(T2DM)in 336 patiants as the research objects,who were hospitalized in Yunnan Province Second People’s Hospital endocrine department between June 2014 and October 2016.The type 2 diabetic patients will be divided into non-retinopathy group(NDR group)142 cases;non-proliferating retinopathy group(NPDR group)70 cases and proliferating retinopathy group(PDR group)124 cases;in addition,taking 72 cases of regular physical examination for normal control group(N group)in our hospital medical center at the same time.Record the clinical data,laboratory test of the biochemical indexes were made.The frequencies of GSTTl-null and GSTP1 genotypes(313A/G)in type 2 diabetic and type 2 diabetic retinopathy and control groups were ascertained by using polymerase chain reaction(PCR)and multiple-PCR techniques.Chi-squqre test and Logistics regression of SPSS 13.0 were used for analyzing the association of the polymorphisms with risk for T2DM and DR.Results:1.There were no statistical differences between N group and T2DM group in alleles(P=0.520;OR=0.85[0.52-1.40])and genotype(P=0.24)of A/G gene of GSTP1 genotype 105#codon;2.There were no statistical differences between NDR group and DR group in alleles(P=0.31;OR=1.26[0.80-1.97])and genotype(P=0.60)of A/G gene of GSTP1 genotype 105#codon;3.Binary Logistic regression analysis confirmed that:there was no correlation between A/G gene of GSTP1 genotype 105#codon and type 2 diabetes and diabetic retinopathy;4.There were statistically significant differences between type 2 diabetes group and the control group in distribution frequency of GSTT1(-)genotype(P<0.05).5.There were statistically significant differences between diabetic retinopathy group and the control group in distribution frequency of GSTT1(-)genotype(P<0.05).6.There were statistically significant differences between type 2 diabetes group and the control group in distribution frequency of TT1(-)AG combined genotype(P=0.02<0.05).7.Correlation analysis between DR and each group of indicators and multivariate regression analysis showed that cardiovascular disease,duration of diabetes and DR were positively correlated(p<0.05),which were risk factors.Conclusion:1.GSTT1(-)genotype may be related to the occurrence of T2DM in Yunnan Han nationality;2.The genetic polymorphism of GSTT1 is not associated with the DR in Yunnan Han nationality;3.There is no correlation between the polymorphism of GSTP1 genotype 105#codon and the occurrence of T2DM and DR in Yunnan Han nationality;4.Individuals with GSTT1(-),GSTP1*AG genotype have a higher risk of developing T2DM;5.There is no significant correlation between GSTT1(-),GSTP 1(313A/G)mutant genotype and retinal lesions,and there is no synergy.6.The occurrence of DR is associated with cardiovascular disease and duration of diabetes. |
