| Background Candida albicans(C.albicans),is the human skin,mucous membranes and digestive tract of the normal flora,is also the most common conditions of human pathogens.When the body’s immune system is damaged or the balance of bacteria is destroyed,Candida albicans is converted into opportunistic killer.More and more studies,especially animal model experiments,show that pattern recognition receptors play a key role in the process of Candida albicans infection.Such as mannan-binding lectin(MBL).MBL acts as a plasma protein produced by the liver,which not only plays an immune defense role in natural immunity,but also participates in acquired immunoregulation.MBL plays a regulatory role in C.albicans phase transformation and its induced immune response.Th17 cells play an important role in C.albicans infection response type,response intensity,duration and so on.Treg cells act as immunoregulatory CD4+T cell subsets that play an important role in the regulation of immune intensity and pathologic damage in anti-C.albicans infection.Is there a correlation between MBL and Th17 and Treg expression in C.albicans infection? What role do MBL play between them? You need to further study.Objective 1.This was aimed to determine the expression of MBL,Th17 cells and Treg cells in the serum of C.albicans infected patients and their relationship with each other.2.Effects of MBL on Treg / Th17 in C.albicans Infected Mice.Methods 1.50 cases of clinical specimens(25 cases of Candida albicans infection and 25 healthy people)were collected and collected according to the inclusion criteria of C.albicans infection.The balance of the two groups was detected.2.Flow cytometry analysis of Th17 and Treg cells: Flow cytometry were used to analyze the changes of Th17 and Treg cells in PBMC of C.albicans infected patients and healthy controls.3.ELISA to detect the levels of MBL and IL-1β: ELISA was used to detect the levels of MBL and IL-1β in peripheral blood of C.albicans-infected patients and healthy controls.4.Th17 and Treg cells were detected by flow cytometry: 4 MBL-/-mice and WT mice were taken,respectively.The Th17 and Treg cells were detected by flow cytometry,and compared with the changes in MBL-/-mice and WT mice.5.Flow cytometry analysis of Th17,Treg cells: C.albicans infected MBL-/-mice and WT mice were 4,flow cytometry detection of Th17,Treg cell ratio.6.Statistical analysis: The experimental data were analyzed by SPSS 21.0 statistical software t-test statistical analysis,P <0.05 that the difference was statistically significant.Results 1.The proportion of CD4+CD25+Foxp3+Treg cells in peripheral blood PBMCs of patients infected with C.albicans increased;and the proportion of CD4+RORγt+Th17 cells also increased.2.The concentration of MBL in serum of patients infected with C.albicans was significantly higher than that in healthy controls.3.Compared with the Kang control group,C.albicans infection in patients with serum levels of inflammatory cytokines IL-1β significantly increased.4.MBL-/-mice Th17 cells were significantly increased,but Treg cells were significantly decreased.5.The proportion of CD4+RORγt+Th17 cells in the MBL-/-mice infected with C.albicans was increased and the proportion of CD4+Foxp3+Treg cells decreased.Conclusion 1.The expression level of MBL in C.albicans infected patients was correlated with the expression of Th17 cells and Treg cells.2.Animal experiments further show that MBL upregulates the expression of Treg,but plays an important role in the expression of Th17. |
