ObjectiveBy RT-PCR to detect the mRNA expression levels of XPC,ERCC1 and XPF in hepatocellular carcinoma (HCC) .Analysising the correlation of the mRNA expression levels of XPC,ERCC1 and XPF and their relationship with clinical pathological parameters and the time of recurrence after operative to discusse the clinical significance of the expression and the role of progression and prognosis in HCC.MethodsXPC,ERCC1 and XPF mRNA expression levels of 73 cases HCC patients and 21 cases of peritumoral tissues were detected by RT-PCR method after the total RNA of the samples were detected by electrophoresis. W-B method was used to detect the protein exprexxion of 10 cases HCC tissues and corresponding peritumoral tissues. All patients did not accept radiotherapy and chemotherapy, interventional therapy and molecular targeted therapy before and after operation.SPSS 17.0 statistical software was applied to analyze the difference of XPC,ERCC1 and XPF mRNA expression in cancer tissues and peritumoral tissues, and the relationship with the clinical pathological features and the recurrence time.A p-value less than 0.05 was condidered significantly.Results1. The total RNA of the samples was degraded in different degrees,but they could still be used for the follow-RT-PCR detection.2. XPC,ERCC1 and XPF amplification curve was significantly "S"type, the exponential growth period was basically parallel,and the melt curve showed a single peak,which showed that the target gene amplification efficiency was similar, and the specificity was well.3. The mRNA expression levels of XPC and XPF was significantly lower than peritumoral tissues (P< 0.05).The mRNA expression levels of ERCC1 in HCC tissues was lower than peritumoral tissues, but the difference was not statistically significant (P>0.05).4. The mRNA expression levels of XPC in HCC were closely related to the liver cirrhosis (P < 0.05). But with other clinical pathological characteristics, there was no significant difference. The XPC mRNA expression levels within different recurrence time groups were significant difference.5. There was a certain correlation between the mRNA expression levels of ERCC1 and membrane invasion.The patiens with membrane invasion haved the lower expression levels of ERCC1 mRNA.6. The mRNA expression levels of XPF were closely related to the levels of ALT before operation, tumor size, membrane invasion and TNM stage.The XPF mRNA expression levels within different recurrence time groups were significant difference (P < 0.05).7. The mRNA expression levels of XPC and ERCC1, XPC and XPF or ERCC1 and XPF were positively correlated in HCC tissues.8. , There was no significant difference in the recurrence rate of 2 years after operation between XPC, ERCC1 and XPF high expression group and low expression group (P=0.866). The recurrence rate of 2 years after operation among high expression of XPC and ERCC1 and low expression of XPF group,high expression of XPC and XPF and low expression of ERCC1 group and high expression of ERCC1 and XPF and low expression of XPC group were not statistically significant difference (P = 0.013)Conclusion1. The mRNA expression levels of XPC and XPF in HCC tissues and peritumoral tissues were significantly different. The mRNA expression levels of XPC and XPF in HCC was significantly lower than the peritumoral tissues. The mRNA expression levels of ERCC1 in HCC tissues were lower.These three genes were expressed at the protein level.2. The mRNA expression levels of XPC in HCC were closely related to the liver cirrhosis. There was a certain correlation between the mRNA expression of ERCC1 and membrane invasion. The mRNA expression levels of XPF were closely related to the ALT levels before operation, tumor size,membrane invasion and TNM stage. The mRNA expression levels of XPC and XPF were related to the time of recurrence after operation.3. The mRNA expression levels of XPC, ERCC1 and XPF in HCC tissues were consistently decreased.4. The combined expression status of XPC, ERCC1 and XPF may be related to the recurrence within 2 years after operation.
