| Purpose:To investigate the effect and therapeutic mechanism of Pingchuang decoction by analyzing its implication for the expression of TNF-α、IL-6 and VEGF in the rat’s oral ulceration models.Methods:Forty-eight rats were divided into normal control group,model group for authenticating,model group,Pingchuang high dose group,Pingchuang low dose group and positive drug group,8 rats in each group,except the normal control group,the rats in the other 5 groups were used to induce rat oral ulceration models by burning with Acetic acid.On second day after the model was made,the ulcer of the rats in the model group for authenticating was observed by eyes,and the oral mucosa tissues were taken for HE staining to observe the pathological changes of the ulcer to test if the oral ulceration model was induced successfully.In the other groups,the rats in the normal group and the model group were treated with distilled water,and the treatment group was treated with the corresponding drugs.After continuous administration for 2 weeks,the general situation of rats and the changes of oral mucosa tissue were observed.The serum levels of TNF-a,IL-6 and VEGF were detected by enzyme linked immunosorbent assay(ELISA).Result:1.General observation:The normal control group rats without oral oral ulcer,mucous membrane is pink,no mucosal edema,no pseudomembrane formation,the rest of the rats after modeling for second days,the right buccal apparent oral ulcer,showed depression,yellow white surface,the surrounding mucosa hyperemia and edema obviously.With the passage of time,the model rat oral ulcer area has a certain degree of reduction,high dose of Chinese medicine group and positive control group rat ulcer ulcer area significantly reduced than before treatment,almost healed,changes of low dose traditional Chinese medicine group rats had no ulcer area Chinese medicine high dose group and positive drug control group,but the group is better than model healing ulcer.2.The result of HE staining:In the model group for authenticating,the oral mucosa of the rats was damaged,the epithelial tissue was broken,the nuclei were broken,ulcers have a large number of inflammatory cell infiltration and local pathological changes were similar to human ROU,the oral ulcer model was copied successfully.In the model group,the oral mucosa was damaged,the epithelial tissue was broken,the nuclei were broken,and the cells were stained and stained deeply.There were a large number of vacuolar degeneration.In the high dose group and the positive control group,the epithelium of the mucosa was more complete,the inflammatory cell infiltration was decreased,and the inflammatory cell infiltration in the low dose group was also improved,but there was no significant difference in the high dose group.3.Weight change:There was no significant difference between the groups.4.The change of ulcer area:the area of oral ulcer in each treatment group was significantly smaller than that in the model group(P<0.01).There was no significant difference between the high dose group and the positive control group.5.The results of ELISA:Compared with the normal group,the serum TNF-a level in the model group and the model group was significantly higher than that in the normal group(P<0.01),and the level of TNF-a was significantly lower in the model group(P<0.05),Compared with the model group,the content of TNF-a in each treatment group was significantly lower than that in the model group,and the content of TNF-a was significantly lower than that of the model group(P<0.01),And compared with the normal control group,although slightly higher than the normal control group,but the difference was not statistically significant(P>0.05),Chinese medicine high dose group and positive control group TNF-a content difference was not statistically significant(P>0.05).Compared with the normal control group,the serum IL-6 levels in the model group and the model group were significantly higher than those in the control group(P<0.01).The level of IL-6 in the model group was significantly lower than that in the model group(P<0.05),Low dose of Chinese medicine group IL-6 compared with that in the model group decreased,the difference was not statistically significant(P>0.05),high dose of Chinese medicine group and positive control group IL-6 compared with that in the model group decreased,the difference was statistically significant(P<0.05),there was no significant difference between Chinese traditional medicine high dose group and positive control group IL-6 content(P>0.05),IL-6 the content of these two groups close to the level of normal control group and normal control group,no significant difference(P>0.05).The content of VEGF in serum of model group and model group for authenticating was significantly higher than that of normal control group(P<0.01),and the difference was significant.Compared with the model group,the content of serum VEGF in the low dose group,high dose group and positive control group were lower than those in the model group,but the difference was not statistically significant(P>0.05).There was significant difference between the high dose group and the positive control group than in the model group(P<0.05).There was no significant difference in serum VEGF level between the high dose group and the positive control group(P>0.05).Conclusion:Pingchuang can effectively reduce the content of TNF-a,IL-6,regulate the expression of VEGF,inhibit inflammation and promote the healing of oral ulcer. |
