| Eupatorium fortunei is the dry and upper part of Compositae Eupatorium fortunei Turez.It is not only used as traditional Chinese medicine,but also ate as food.According to Chinese medicine theory,its function ranges from removing dampness by means of aromatics,activating spleen to expelling summer-heat.This project about Eupatorium fortunei is based on the national scientific research of traditional Chinese medicine industry:key technical specifications of producing and processing in 30 kinds of Chinese herbal medicine(201507002).Its aim is to solve the problems encountered in Eupatorium fortunei’s processing and producing process,such as no uniformed processing method,no reproducible technology,no scientific statistical support.This thesis is divided in the following 5 parts:1.Literature researchIn this part,the chemical constituents and pharmacological effects of Eupatorium fortunei are systematically summarized.At the same time,the history,advantages and implementations of the integration of producing and processing of Chinese medicine are summarized.2.Original investigation of Eupatorium fortuneiIn this part,the main producing areas of Eupatorium fortunei in history and also modern times are visited.Environmental conditions,planting area,cultivation methods,harvest time,processing and preservation methods of four main production areas(Sichuan,Jiangsu,Anhui and Henan)are analyzed and summarized.3.Research on integrated processing production and technology of Eupatorium fortune.We selected 5 kinds of volatile components:1-Methyl-3-methoxy-4-isospropylbenzene,thymol,and caryophyllene oxide,β-caryophyllene,and a-caryophyllene as targets to be simultaneously determined by GC-MS.The effects of processing equipment and temperature on these components were also investigated.4.Quality evaluation of different processing methods for Eupatorium fortune.Then,a pilot test was conducted at Shanghai Edward factory to compare the differences between traditional producing method and the optimized method.Extraction and determination of total flavonoids:BBD response surface method combined with PB test were used to build the optimum extraction method of flavonoids from Eupatorium fortunei.The content of total flavonoids was determined by UV spectrophotometer.Establishment of HPLC fingerprint:HPLC-UV method was used to establish the fingerprint of Eupatorium fortune.Coumarin is used as the reference peak.5.Quality control of Eupatorium jortune for processed pieces and integrated processed pieces.Determination of inorganic elements:using Inductively Coupled Plasma-Mass Spectrometry(ICP-MS)to determine 13 inorganic elements(As,Cr,Cu,Ga,Ba,Cd,Mn,Ni,Pb,Sb,Sn,Sr,T1)in the samples coming from 21 areas and commodity specifications.The principal component analysis and correlation analysis were performed by using SPSS software.Research on HPLC fingerprint:The fingerprints of commercial herbs,crude drugs and decoction were studied.Innovation point:1.By comparing the content of volatile oil and total flavonoids,we determined that the quality of half year Eupatorium fortunei was better than that of one year.2.Apply the optimized method to the production in factory.Determine the effects of the drying time before Eupatorium fortunei cutting and cut length on the quality of product. |
