ObjectiveTo observe the effects of Suhuang Zhike capsules on a guinea pig model of cough variant asthma.Methods50 female guinea pigs were randomly divided into five groups:normal control group,model control group,prednisone acetate tablets group and Suhuang Zhike capsules high and low doses group,combined with the related literatures to set up the guinea pig model of cough variant asthma.Fumigation was performed in half an hour for model control group,prednisone acetate tablets group,Suhuang Zhike capsules high and low doses group each day from day 1 to day 29.At day 15 and 22,the suspension containing 1ml OVA(2%)and 200mg AL(OH)3 was administered by intraperitoneal injection to the guinea pigs.Atomization attack was carried out by 1%OVA from day 29 once each day,for 7 days.The administration began on the day of the nebulization challenge on day 30,the dose of Suhuang Zhike Capsule was 6g/kg in the high dose group,2g/kg in the low dose group and 3mg/kg in the prednisone acetate group.Gavages to give the corresponding drugs,once a day for 7d.Normal control group and model control group were given the same dose of saline.The changes of coughing times induced by capsaicine were monitored at day 35.The changes of airway resistance were observed at day 36 and eosnophils in bronchoalveolar lavage fluid were further monitored,and the serum IL-4,IFN-γ,TNF-α,and IgE levels.ResultsCompared with the normal control group,the cough frequency and the percentage by eosinophils in BALF of the model control group was significantly increased(P<0.05),the resistance in guinea pig of model control group in methacholine concentration by 0.2mg/ml,0.4mg/ml was increased obviously(P<0.05),the serum IL-4,TNF-y and IgE levels of model control group increased significantly(P<0.05).Cough times measurement,compared with the model control group,the c ough frequency of Suhuang Zhike capsules high doses group was reduced obv iously(P<0.05);Compared with the Suhuang Zhike capsules low dose group,the cough frequency of Suhuang Zhike capsules high doses group was reduce d obviously(P<0.05).Airway resistance measurement,the resistance in guinea pig of prednisone acetate tablets group in methacholine concentration by 0.2mg/ml,0.4mg/ml was increased obviously(P<0.05);The resistance in guinea pig of Suhuang Zhike capsules high dose group in methacholine concentration by 0.2mg/ml,0.4mg/ml was decreased(P<0.05),the resistance in guinea pig of Suhuang Zhike capsules low dose group in methacholine concentration by 0.2mg/ml was decreased(P<0.05);Compared with the resistance in guinea pig of prednisone acetate tablets group and Suhuang Zhike capsules low dose group in methacholine concentration at 0.2mg/ml and 0.4mg/ml,Suhuang Zhike capsules high dose group was decreased obviously(P<0.05).Bronchoalveolar lavage fluid(BALF),compared with the model control group,the percentage by eosinophils in BALF of the three treatment group was decreased(P<0.05),the total white blood cells of Suhuang Zhike capsules high dose group was decreased(P<0.05);Compared with the Suhuang Zhike capsules low dose group and prednisone acetate tablets group,the total white blood cells of Suhuang Zhike capsules high dose group was decreased obviously(P<0.05);Compared with the Suhuang Zhike capsules low dose group,the percentage by eosinophils in BALF of the prednisone acetate tablets group was decreased obviously(P<0.05).Determination Of IL-4、IFN-γ、TNF-α and IgE in the serum,compared with the model control group,the serum IL-4 level of prednisone acetate tabletsgroup was significantly decreased,and the serum TNF-α and IgE of Suhuang Zhike Capsules high dose group decreased significantly(P<0.05);Compared with the Suhuang Zhike Capsules high dose group,the serum IL-4 level of prednisone acetate tabletsgroup was significantly decreased(P<0.05);Compared with the Suhuang Zhike Capsules low dose group,the serum IgE level of Suhuang Zhike Capsules high dose group was significantly decreased(P<0.05).ConclusionSuhuang Zhike capsules could reduce serum TNF-α levels,ameliorate the cough frequency,airway inflammation and resistance in CVA guinea pig,so as to achieve the purpose of the treatment of CVA.
