Purpose: Analysis on the correlation between hypertension candidate gene polymorphisms and hyperactivity of liver Yang of hypertension patients.Method:The study choose 60 hypertension patients from department of cardiology from inpatient department and specialist outpatient treatment in Rui Kang Hospital Affiliated of Guang Xi University of Chinese Medicine and The First Affiliated Hospital of Guang Xi University of Chinese Medicine from December 2015 to December 2016 as the research object,and collect patients clinical symptoms and signs within one month,the history of hypertension,medication within two weeks,complication,family history of hypertension through the establishment of the registration form for patients with hypertension and questionnaires;According to the traditional Chinese medicine theory,we establish registration form of the type of traditional Chinese syndromes;qualified subjects are divided into three groups,namely the normal blood pressure group,hypertension of liver yang hyperactivity group,non-hypertension of liver yang hyperactivity group,and 30 cases from each group are intended to study;The venous blood are all extracted in the median cubital to extract DNA.DNA is extracted from peripheral blood leukocytes by saturated salting out method,and polymerase chain reaction(PCR)is combined with restriction endonuclease method for detecting gene polymorphism;We collect and test the type of hyperactivity of liver Yang from the hypertension TCM syndromes type,genotype and gene frequency analysis by SPSS14.0software.Enumeration data is by t test,and measurement data is by x~2 test.The result is objectively summarized and evaluated.Results:(1)the basic situation of the object of study: The liver yang hyperactivity group and non-liver yang hyperactivity group are compared with normal control group on gender,age and body mass index and heart rate,and it can been seen that the difference is not statistically significant(P>0.05);The systolic blood pressure,diastolic blood pressure,CHOL level,HDL-C level,LDL-C level,Lp A level,pulse pressure,ACE,AGT,Ang II of liver yang hyperactivity group and non-liver yang hyperactivity group are close and there is no obvious difference;but it increases significantly when it is compared with the control group,and there is significant difference(P<0.05).(2)risk factors: The rate of smoking,drinking,type 2 diabetes,coronary heart disease,carotid plaque about hyperactivity of liver Yang Group and the non-liver yang hyperactivity group is significantly higher than that of the control group,compared with the control group,there is significant difference between the two groups(P<0.05).(3)Three ACE genotypes are divided into: DD type,ID type,II type and allele frequency is D type and I type;for liver yang hyperactivity group and non-liver yang hyperactivity group,each genotype and allele frequency of P are all more than 0.05,and there is no significant statistical difference between groups;genotype and allele frequencies of hyperactivity of liver Yang and the non-liver yang hyperactivity group are compared with control group respectively,and P are all less than 0.05,and there is significant statistical difference between groups.(4)Three ACE genotypes are divided into: MM type,MT type,TT type and allele frequency is M type and T type;the comparison between liver yang hyperactivity group,non-liver yang hyperactivity group,and control group is that each genotype and allele frequency P are all less than 0.05,and there is significant statistical difference between groups.(5)CHOL,TG,HDL-C,LDL-C,Lp A,ACE,AGT,Ang Ⅱof liver yang hyperactivity group are compared in DD type,ID type,II type of the ACE and the difference is statistically significant(all P <0.05).The indicators of the non-liver yang hyperactivity group are compared in DD type,ID type,II type of the ACE and the difference is statistically significant(all P <0.05).(6)CHOL,TG,HDL-C,LDL-C,Lp A,ACE,AGT,Ang Ⅱof liver yang hyperactivity group are compared in MM type,MT type,TT type of the ACE and the difference is statistically significant(all P <0.05).The indicators of the non-liver yang hyperactivity group are compared in MM type,MT type,TT type of the ACE and the difference is statistically significant(all P <0.05).Conclusion:(1)ACE,AGT,AngII level of patients with hypertension is associated with hypertension morbidity,and has a certain relevance of hyperactivity of liver Yang,but the specific mechanism is unknown.(2)There is no significant difference for comparison between liver yang hyperactivity group and non-liver yang hyperactivity group for each genotype of ACE I/D site and allele frequency.There is significant difference for comparison between liver yang hyperactivity group and control group.There is also significant difference for comparison between non-liver yang hyperactivity group and control group.It can only show that each genotype of ACE and I/D site may be associated with hypertension,but it still cannot show that each genotype of this site is associated with hyperactivity of liver Yang of hypertension.(3)There is no significant difference for comparison between liver yang hyperactivity group and non-liver yang hyperactivity group for each genotype of AGT M235 T site and allele frequency.There is significant difference for comparison between liver yang hyperactivity group and control group.There is also significant difference for comparison between non-liver yang hyperactivity group and control group.It can show that each genotype of AGT M235 T site may be associated with hyperactivity of liver Yang of hypertension.(4)ACE I/D site has no significant correlation to TCM syndrome types of hypertension;AGT M235 T site may be an important risk of primary hypertension of hyperactivity of liver Yang,which has a certain reference value for preliminary screening in hypertension susceptibility genes. |
