Polygala. tenuifolia Willd. is commonly used herbs, it has long been documented in "Shen Nong’s Materia Medica", it is widely used in clinical practice because it has a good effect on the nerves puzzle, expectorant swelling. With the gradual expansion of Chinese herbal medicines demand, Polygala artificial cultivation technology has evolved in geoherb and its surrounding areas, but the problems of low seed germination rate, insufficient level of cultivation techniques, high yield and poor quality still exist. The study found that secondary metabolites under stress conditions tend to be more conducive to the accumulation, but manufacturing adversity environment does not have the maneuverability in the large-scale cultivation of P. tenuifolia Willd., and long-term, excessive stress make the herbs negative growth. In recent years, studies have found that the closely relationship between development and secondary metabolites accumulation of salicylic acid(SA) and plant growth hormone. By the way of spraying SA, the key problems of P. tenuifolia Willd. cultivation research include:exploring the biosynthetic pathway and accumulation rules of P. tenuifolia Willd. medicinal ingredient. The experiment investigate the effect of seed soaking with SA on germination of P. tenuifolia Willd., explore the effect of spraying SA on polygalaceae seedling growth, physiological and biochemical accumulation and pharmaceutical active ingredients, analyse the relationship between the concentration of salicylic acid spraying and applying different time for different medicinal ingredient accumulation. The main results are as follows:1. With the increase of storage years, P. tenuifolia Willd. seeds membrane lipid peroxidation will deepen, MDA content will increase, vitality of antioxidant enzymes such as CAT and POD will decrease, germination rate, germination energy, germination index will decrease. Different concentration levels of SA, GA, MeJA treat P. tenuifolia Willd. old seed, we has found the germination of different concentration SA can significantly improve the old seeds of P. tenuifolia Willd., and the effect of 50mg/L SA is best,50mg/L and 100mg/L GA can also enhance the effect of germination; MeJA is not suitable for seed soaking of P. tenuifolia Willd., Each concentration of MeJA can inhibit the germination rate, germination energy and germination index.2.0.15 mmol/L SA for Polygala tenuifolia aboveground and root biomass accumulation and content of UV-absorbing material have a facilitating role. High concentration (5 mmol/L) SA for the entire growth period of P. tenuifolia Willd. anthocyanins, carotenoids promote accumulation remains high, but SA (5 mmol/L) has a long-term inhibition of the accumulation of UV-absorbing compounds. The concentration of SA is lower than 1.5 mmol/L can inhibit the accumulation of MDA, High concentration of SA will continue to promote the accumulation of MDA. SA will increase CAT and POD activity, with the growth time prolonged, the inhibition of SA on POD will appear. P. tenuifolia Willd. leaves after being stimulated by exogenous SA, PPO activity increase, PAL activity in the short term has been on the rise, but SA is the stage for the induction of PAL, the most lasting 20 days.3. Effect of SA on the aerial parts as follows:0.5 mmol/L SA treatment is favorable to accumulate the total phenolics(PNL) and mangiferin(MFN) of aerial parts P. tenuifolia Willd.; 0.15 mmol/L SA treatment is favorable to accumulate the total flavonoids(XTE) and total saponins(ONS) of aerial parts P. tenuifolia Willd.; 5 mmol/L SA treatment is favorable to accumulate the 3,6’-disinapoyl sucrose(DISS) of aerial parts P. tenuifolia Willd.. Effect of SA on the roots as follows:0.5 mmol/L SA treatment is favorable to accumulate the PNL, ONS, DISS and polygalaxanthone Ⅲ(PXT) of roots P. tenuifolia Willd.; 0.05 mmol/L SA treatment is favorable to accumulate the XTE of roots P. tenuifolia Willd.; 5 mmol/L SA treatment is favorable to accumulate the MFN of roots P. tenuifolia Willd.. Comprehensive consideration, the quality of pharmacodynamics parts are improved significantly by spraying 0.5 mmol/L SA.4. Using different concentrations SA to stimulate P. tenuifolia Willd. Samples and collecting samples by segmenting time make the physiological and biochemical index and pharmacological ingredients appear significantly different, then the relationship of physiological and biochemical index and pharmacological ingredients is analyzed. The results show that:PAL and PNL, XTE, ONS, MFN, PXT have a significant positive correlation, PAL and DISS have a significant negative correlation; POD and DISS have a significant positive correlation, POD and all other medicinal ingredient have a significant negative correlation; Photosynthetic pigments and aerial parts DISS, roots DISS have a significant positive correlation, photosynthetic pigments and PNL, XTE have a significant negative correlation.
