Knee osteoarthritis(KOA)is a degenerative bone and Joint disease characterized by osteoarticular cartilage degeneration,subchondral bone reconstruction(thickening,bone atrophy,bone cyst),ligament and meniscus degeneration,joint capsule hypertrophy.It seriously damages to human health,restricts population labor and affects the quality of life in the middle age,resulting in social and family burden.Western medicine think that its pathogenesis is unclear.Western medicine treatment,include drugs,physical therapy,brace,exercise therapy and surgery.but there is no satisfactory treatment.Jian Gu recipe that is Hubei Chinese medicine master Professor Li Tongsheng prescription,can promote to repair of articular surface,and achieve good results in early and mid-KOA’s treatment.The experimental results of early rabbits showed that Jiangu recipe could relieve arthritis and improve articular cartilage function,but its exact mechanism should be studied deeply.This paper explore the mechanism of Jiangu recipe on p38 MAPK signal transduction in the treatment of delayed articular cartilage degeneration,dealing with the expression of p38 MAPK and the degradation of type Ⅰ andⅡ collagen in the treatment of KOA.OBJECTIVE: To observe the effect of Jian Gu recipe on the articular cartilage,The effect of Jiangu recipe on the expression of type Ⅰ and Ⅱ collagen i,and the effect of KOA on p38 mitogen-activated protein Kinase(p38 MAPK)expression in KOA model.Methods: Male and female rats were randomly divided into four groups: normal group,(Sham operation group treated physiological saline).The model group(Surgery group treated physiological saline),the control group(Surgery group treated celecoxib).Jiangu recipe group(Surgery group treated Jiangu recipe).After 3 months of treatment,the rats were sacrificed.Observe the morphological changes of articular cartilage under microscope.Detect the expression of type Ⅰ and Ⅱ collagen m RNA in articular chondrocytes by RT-PCR.Detect the expression of p38 in articular chondrocytes by Western blot,to observe the effect of Jiangu Tang on protein kinase in p38 MAPK signal transduction pathway.Results:we found that the model group articular cartilage tissue red staining showed necrosis,from electron microscopy results.While Jiangu recipe group articular cartilage is rough,formate of ulcers,exist cartilage matrix in a small amount of necrotic area,not liking the model group.Pathological changes in Mankin,s score results showed that the model group and the normal group,the healthy bone group and the model group were significantly different(p<0.05,p=0.000);Jiangu recipe can reduce the expression of type I collagen m RNA and increase the expression of type II collagen in articular chondrocytes;P38 MAPK phosphorylation products were expressed in all groups.That p<0.05 in the model group compared with other groups explains the pathogenesis of osteoarthritis is inducedby the activation of p38 mitogen activated protein kinase signal transduction pathway.The expression of phosphorylated p38 MAPK decreased,compared with the model group.Conclusion: The prescription of Jiangu recipe has obvious effect on the repair of articular cartilage of knee osteoarthritis rat model induced by Hulth.The pathogenesis of knee osteoarthritis may be closely related to p38 MAPK mediated cell signaling pathway.Through the intervention and regulation of p38 expression and activation of mitogen activated protein kinase signal transduction signal,will hopefully become a new effective way for the treatment of knee osteoarthritis. |
