Objective: To study the effect of TAN influenced the proliferation ability and freeradicals of TNF-a-induced osteoarthritis model chondrocytes cultured in vitro. Toargument the protect mechanism of TAN to chondrocytes, and explore the clinicalusage scope of TAN.Methods: Chondrocytes from the knee joints of 4-weeks-old white rabbite werecultured in vitro in 24-Multiwell, then were used for evalution of proteoglycan andcollagenⅡwith AB-PAS staining, biochemical analysis, immunohisyochemistry andtransmission microscopy. The chondrocytes were divided into 8 groups. Controlgroup was normal chondrocytes. Experimental group was mixed into TNF-a. Exceptfor experimental group and control group, other groups were mixed into differentdensity TAN and TNF-a. After 48 hours, MTT method examed the proliferationability of chondrocytes. Two density TAN groups were selected for next step.Chondtocytes were divided into 4 groups, the control group was not mixed intoanything, the experiment group were mixed into TNF-α, other groups were mixedinto TNF-a and two density TAN. After 72 hours, observe the change of SOD andiNOS.Results: 1 Immunohistochemistry revealed that there was a staining of collagenⅡ.2 5μg/ml TAN group, 7.5μg/ml TAN group and 10μg/ml TAN group could improveproliferation ability, and had more significant statistics different than the experimentalgroup. While 15μg/ml TAN group,20μg/ml TAN group and 50μg/ml TAN group hadno statistics different compared with experimental group. 3 5μg/ml TAN group couldenhance SOD activity, and had significant statistics different compared with controlgroup. SOD activity of 10μg/ml TAN group had statistics different compared withexperimental group, but there was no statistics different between 10μg/ml TAN groupand control group. 4 There was statistics different between control group and experimental group. 5μg/ml TAN group could not decrease the iNOS activity. 10μg/mlTAN group could decrease the iNOS activity and had significant statistics differentcompared with control group.Conclusion: 1 TAN could enhance chondrocytes proliferation ability.2 TAN could enhance SOD activity and increase iNOS activity.
