| Objective: To observe the Longzuan Tongbi Recipe of Guangxi Zhuang medicine on rheumatoid arthritis(ra)model in rat serum cytokine interleukin-6 and synovial toll-like receptors in 4,the influence of myeloid differentiation factor 88,discusses on the zhuang medicine dragon drill through polioplus partners party TLR4 / NF-K B mechanism of signal transduction pathways,for the party certification of zhuang medicine dragon drill through provides the basis for clinical application.Methods:through the establishment of a rat model of adjuvant induced rheumatoid arthritis,divided into blank group,model group,positive control group,Longzuan Tongbi Recipe of Guangxi Zhuang medicine high dose,middle dose and low dose group.Record the weight and diameter of ankle joint of rats changes,observe the influence of Longzuan Tongbi Recipe of Guangxi Zhuang medicine rat model for foot swelling and weight changes,and the use of inflammatory factor IL-6 enzyme linked immunosorbent assay in rat serum,using reverse transcriptase polymerase chain reaction to detect TLR4 and MYD88 gene expression in synovium of rats the situation,using immunohistochemical method to detect the synovium of rats and the expression of MYD88 protein TLR4.Results: Longzuan Tongbi Recipe can make rheumatoid arthritis rats weight gain in the model group,high dose group weight gain than the low dose group,including dose group compared with model group can reduce rats ankle diameter.The comparison of the high dosage group and the medium dose group in the high dosage group and the middle dose group of the Longzuan Tongbi Recipe of Guangxi Zhuang medicine reduced the levels of il-6 in the serum of the model rat.Compared with the model group rats,the high dose and the medium dose group of the strong drug were reduced by the mRNA expression of TLR4 and MYD88 in the synovial tissue.Immunohistochemical slices,the cell membrane is brown color(yellow)is positive cells,and based on the staining intensity of positive cells and the ratio of the semi-quantitative score,Longzuan Tongbi Recipe of Guangxi Zhuang medicine parties including the dose group of immunohistochemical score was lower than those of model group rats.Conclusion: Longzuan Tongbi Recipe can increase the weight of rats,shorten the ankle diameter of rats,improve the inflammatory response in rats,regulate inflammatory cytokines IL-6,reduce the TLR4 and MYD88 gene and protein expression of zhuang medicine dragon tong bi side is a relatively effective anti-rheumatism medicine,its mechanism may be related to by adjusting the TLR4 / NF-kB signal transduction network and related factors. |
