Lumbar Intervertebral Between TCM Syndrome Type And Degeneration Of Intervertebral Disc Tissue Of MMP-2 And MMP-13 Correlation Studies

Objective:observation of patients with lumbar disc prolapse 1,professional characteristics,in terms of gender,age,body weight,degree of intervertebral disc degeneration Pfirrmann grading standard,pain degree and the properties index and the correlation of TCM syndrome types;Comparison between different syndrome type 2 disc nucleus pulposus tissue of MMP-2,expression of MMP-13 if there is a difference,3 through the detection of MMP-2 and MMP-13 in the intervertebral disc nucleus pulposus tissue expression,analysis of correlation between the two is discussed in this paper.Methods: 1.Clinical research According to the diagnosis of lumbar disc prolapse,TCM syndrome type classification standard survey plan.From April 2016 during an April 2017 in XXX for lumbar intervertebral disc protrusion bone second division hospital nucleus pulposus intervertebral foramen mirror line of lumbar intervertebral disc protrusion with excision or simply open the window in patients with excision of nucleus pulposus,reference set by the state administration of traditional Chinese medicine "standard of diagnosis of disease and curative effect of traditional Chinese medicine" as the standard,respectively into the qi and blood stasis group and kidney deficiency syndrome group,20 cases in each group.Survey content includes the general situation of patients,symptoms of TCM characteristics,degree of intervertebral disc degeneration Pfirrmann grading standard,etc.,using modern statistical methods,discusses the objective index and the correlation of TCM syndrome types.2.The experimental research on specimens from the clinical research of the two groups of the patients with nucleus pulposus tissue,respectively into the qi and blood stasis group and kidney deficiency syndrome group,20 cases in each group.The group will be out of the nucleus pulposus and fiber ring immediately after separation with sterile saline wash clean,in a specimen bag,pour into formaldehyde solution impregnation,completely fixed after 24 hours to the laboratory,embedding,sectioning.Using HE staining to observe morphological changes of nucleus pulposus in degeneration,using immunohistochemical method to detect MMP-2 and MMP-13 in the expression of nucleus pulposus in degeneration.Clinical research results: 1.40 cases of patients with lumbar disc prolapse,with qi and blood stasis patients and 20 patients with liver and kidney deficiency syndrome.Two groups of patients with TCM syndrome type of gender,weight,occupation characteristics,P > 0.05,there was no statistically significant difference.Age and course of the disease with TCM syndrome type by the single factor analysis of variance,P < 0.05,the difference was statistically significant.In terms of degree of intervertebral disc degeneration Pfirrmann grading standard,the level of two groups of TCM syndrome type distribution,the nonparametric test,P < 0.05,the difference was statistically significant.In terms of degree of pain,is pain score(VAS score)in the differences between the two groups of TCM syndrome type,the single factor analysis of variance,P < 0.05,there was no statistically significant difference.In the nature of pain,more pain properties in the differences between the two groups of TCM syndrome type,the nonparametric test,P < 0.05,the difference was statistically significant.In daily activity ability,daily activity ability in want to rent the difference between TCM syndrome types,the single factor analysis of variance,P < 0.05,the difference was statistically significant2.Experimental study observed HE staining results nucleus pulposus cells distributed clutter,the nucleus for the dark blue,red,surrounding tissue is arranged disorderly,change of nucleus pulposus cells free bubble sample.Immunohistochemical staining results of MMP 2,MMP-13 in the specimen has a certain degree of expression of MMP-2 and MMP-13 positioning in the cytoplasm of nucleus pulposus,immunohistochemical staining positive immune reaction material dyeing as brown,nuclei are in blue.The expression of MMP-2 of MMP-2 in the group of qi and blood stasis,liver and kidney deficiency,the positive expression rate of respectively(43.07 + 4.01)%,(74.93 + 12.05)%.Two groups between MMP-2 positive expression rate of TCM syndrome types by the single factor analysis of variance,F = 62.974,P = 0.000 < 0.05,the difference was statistically significant.The expression of MMP-13 cases MMP-13 in the group of qi and blood stasis,liver and kidney deficiency,the positive expression rate of respectively(46.75 + 7.34)%,(77.41 + 10.15)%.Two groups between MMP-13 positive expression rate of TCM syndrome types by the single factor analysis of variance,F = 59.950,P = 0.000 < 0.05,there was no statistically significant difference.Degeneration of intervertebral disc nucleus pulposus tissue positive expression of MMP-2 was a positive correlation with the positive expression of MMP-13(r = 0.812,P = 0.000 < 0.05).Conclusion: patients’ gender,body weight,professional characteristics and no obvious correlation between TCM syndrome type lumbar disc prolapse.Age,duration,and degree of intervertebral disc degeneration Pfirrmann grading standards,the nature of the pain,the TCM syndrome types have a certain correlation with lumbar intervertebral disc protrusion,can be used as a reference index of traditional Chinese medicine.The incidence of liver and kidney deficiency type with the extension of the course is on the rise.In terms of degree of intervertebral disc degeneration Pfirrmann grading standard,see more at qi stagnancy and blood stasis syndrome grade II and grade III,liver and kidney deficiency type found in IV,V.In terms of nature of pain,see more at a stabbing pain qi stagnancy and blood stasis syndrome,liver and kidney deficiency type found in pain.Liver and kidney deficiency type group of nucleus pulposus MMP-2 and MMP-13 positive expression rate is significantly higher than that of qi and blood stasis type,can be used as a reference index to distinguish the two kinds of card type,for objectification and standardization of traditional Chinese medicine provides the basis.MMP-2 and MMP-13 in the degenerative intervertebral disc nucleus pulposus tissue have a clear positive correlation,speculated that MMP-2 and MMP-13 May be involved in the process of adjustment.
