Objective To investigate the expression and clinical significances of endocan,VEGF in human brain gliomas and the relationship between endocan and pathlogic grade of gliomas,MVD. Then look for a new marker of glioma clinical diagnosis and treatment.Methods We investigate the expression leves of endocan m RNA with reverse transcription-polymerase chain reaction(RT-PCR)in 51 cases gliomas.Immunohistochemistry was used to detect the expression levels of endocan,VEGF,CD-34 and Ki-67 in 11 cases normal brain and 183 cases gliomas. Their correlation to the pathologic grade,and the relationship between the expression of endocan and MVD in human gliomas were studied.Results 1. The immunohistochemistry stain suggested that in normal brain tissues endocan is expressed in neuron,never expressed in glial cell neither endothelial of vessels.The expression of endocan and VEGF are observed in all pathlogic grade of gliomas. The increasing expression levels of endocan and VEGF had positive correlation with the ascending of pathlogic grade of tumor specimen(P<0.01).2. The immunohistochemistry stain suggested that the expression of CD34 is located in cell membrane. In normal brain tissues, microvasculars with small lumen and same structure are evenly distributed. In low grade of gliomas(WHOⅠ、Ⅱ),there are more microvasculars which exprsse CD34,The most number of CD34 expression in microvascular are observed in high grade gliomas(WHOⅢ、Ⅳ). There was a positive and significant correlation between the expression leves of MVD and the grade of glioma(P<0.01).3. The immunohistochemistry stain suggested that the expression of Ki-67 is located in cell nucleus. There was a positive and significant correlation between the expression leves of Ki-67 and the grade of glioma(P<0.01).4. RT-PCR suggested that,The expression leves of endocan mRNA were significantly higher in high grade gliomas(WHOⅢ Ⅳ) than those in low grade gliomas(WHOⅠ Ⅱ)(P=0.001).5. Correlation analysis suggested that,There was a positive and significant correlation between the expression leves of MVD and endocan as well as VEGF(r=0.55, P<0.01;r=0.64,P<0.01).There were positive and significant correlations between the expression leves of Ki-67 and endocan as well as VEGF(r=0.43, P<0.01;r=0.58,P<0.01).Conclusions The high expression leves of endocan in gliomas means that there may be a higher malignancy and proliferative activity. So we could get that endocan play an important role in predicting the malignancy and proliferation of human gliomas.
