Objective:To investigate the inhibitory effects of anti-Fas hammerhead ribozyme on Fas and Fas mediated apoptosis of T lymphocytes in COPD mice, and to observe the changes of T lymphocytes subsets.Method:Sisty clean-class healthy male mice were chosen and adaptively raised for two weeks.Afterwards they were randomly divided into healthy controls group(ten) and COPD group(fifty),meanwhile COPD mice models were obtained by exposing COPD group to smoke to observe the changes of pathologic slices brought by hematoxylin-eosin(HE) staining and pulmonary function test..After successful modeling,COPD group were devided into group A(copd),group B(vector transfection), group C(anti-Fas hammerhead ribozyme) and group D(mutated ribozyme) randomly.Empty transfection and anti-Fas hammerhead ribozyme and mutated ribozyme were injected into group B and C and D.The Fas expression of T cells was detected by RT-PCR and Western blot to obseve the apoptosis rate of CD4+T cells and the ratio of CD4+and CD8+ were observed by flow cytometry.Results:(1)Compared with healthy controls group, the COPD group had an enhanced inspiratory resistance(RI) with a decline on peak expiratory flow(PEF) and dynamic compliance(Cdyn),the results have statistical significances,P<0.05,and pathologic results can be observed as HE staining showing the alveolar expansions in lung parenchyma,inflammatory cell infiltrations in small airway.these evidences prove that the COPD mice model were built successfully.(2) Compared with group A, the Fas expression of T cells in group B had no significant difference,P>0.05,and the Fas expression of T cells in group C and D were reduced, the results have statistical significances,P<0.05.The Fas expression of T cells in group C was lower than those in group D, this result shows statistical significance,P<0.05.(3) Compared with group A, the apoptosis rate of CD4+T cells and the ratio of CD4+and CD8+had no significant differences in group B,P>0.05.The apoptosis rate of CD+T cells was reduced while the ratio of CD4+and CD8+was increased in group C and D, the results have statistical significances,P<0.05.And compared with group D,the apoptosis rate of CD4+T cells was lower while the ratio of CD4+and CD8+was higher in group C, the result of which has statistical significances,P<0.05.Conclusion: Anti-Fas ribozyme can remarkably inhibit the Fas expression and prevent CD4+T cells from Fas-mediated apoptosis which enhanced their ability against apoptosis and optomized the ratio of CD4+ and CD8+in COPD mice.
