Objective:To explore the effect of Pu Jin oral liquid in the treatment of cerebral palsy By observing TNF-α and IL-6 expression in the brain tissue,and the behavior of related indicators,After giving Pu Jin oral liquid to the ntrauterine infection/ inflammation caused by premature brain injury in rats.Provide new theories,treatment method and effective drugs for the occurrence and development of premature PVL,which was caused by intrauterine infection/inflammation,so as to reduce the incidence of cerebral palsy.Method:(1)Animal model: Thirty pregnant rats were divided into LPS group(n = 24)and the control group(n = 6)randomly.LPS group was injected with LPS solution by intraperitoneal,and the control group was injected with saline in the 16 th and 17 th day of pregnancy,and the injection dose of the two groups was 350ug/kg.Premature rat is pregnancy < 22 days of delivery.Start the uterus and placenta HE staining immediately in mother mouse after parturition,so as to observe intrauterine infection/inflammation.Observe pathological changes after the birth of baby mice by taking brain tissue for HE staining.(2)Animal grouping: 80 preterm pups of LPS group were divided into eight groups by stochastic method,and each group is divided into 10.Early drug intervention group include A1,B1,C1 and D1.Late drug intervention group include A2,B2,C2 and D2.A1 and A2 were low-dose group of Pu Jin Oral Liquid,B1 and B2 were High-dose group of Pu Jin Oral Liquid,ginkgo biloba extract groups were C1 and C2,model groups were D1 and D2.(3)Intervention methods: Early drug intervention group used intragastric administration on the birth day of the rats.Late intervention group used intragastric administration on the 7th day after birth.Intragastric administration of the two groups for seven consecutive days.Raised to 21 days of age under the same conditions after the intervention,then start neuroethology test(use Suspension Test,Ramp Test and Cylinder Test).(4)Nine groups of young rats fed to 21 days under the same conditions,then take brain tissue.Using immunohistochemical method to observe the expression of IL-6 and TNF-α.Results:(1)general situation: LPS group of pregnant rats were prone to miscarriage and premature delivery after modeling,and had die in childbirth happened.The number of childbirth in mice of LPS group of pregnant rats Less than the control group(P <0.05).Premature rat of LPS group had Lower body weight(P < 0.05)compared with the control group,less activity,skin cyanosis and weak stress response.(2)HE dyed pathomorphism changes: Uterine and placental tissue pathological examination of LPS group of pregnant rats can be observed artery congestion and a large number of neutrophil infiltration,but the control group did not see inflammatory reaction.Pathological detection of LPS group early litter mice observed the Structure sparse of the cerebral white matter,corpus callosum and internal capsule,and nerve fibers arranged not neat,and number of neurons was less,and lateral ventricle and cerebral white matter had bleeding,and the glial cell aggregation.(3)neurobehavioral ability: High and low dose Pu Jin oral liquid and ginkgo biloba extract all can improve premature brain damage rats neurobehavioral ability,which compared with model group(P < 0.05),and the effect of early intervention is clearly better than the late intervention.High and low dose Pu Jin oral liquid and ginkgo biloba extract were no significant difference among three groups In improving premature brain injury rats neurobehavioral ability(P>0.05).(4)IL-6 and TNF-α relative expression of results: IL-6 and TNF-α relative expression significantly raised of the LPS group mouse tissues compared with the control group(P < 0.05).In the early drug intervention group,compared with model group,Pu Jin both high and low dose oral liquid and ginkgo biloba extract can significantly lower seed mouse tissues of IL-6 and TNF-α relative expression(P < 0.05).The role of Pu Jin oral liquid high dose downgrade IL-6 and TNF alpha relative expression is obviously better than the low dose and ginkgo biloba extract,in the high and low dose Pu Jin oral liquid and ginkgo biloba extract comparison between three groups(P < 0.05).But Pu Jin oral liquid of low dose and ginkgo biloba extract,which had no significant differences in downgrading IL-6 and TNF alpha relative expression.In the late drug intervention group,high and low dose Pu Jin oral liquid and ginkgo biloba extract all can obviously decrease relative expression,of which both IL-6 and TNF-α in the brain tissue of the mouse.Pu Jin oral liquid high dose downgrade IL-6 and TNF-α relative expression was obviously better than the effect of low dose and ginkgo biloba extract(P < 0.05),the effect of ginkgo biloba extract was more than Pu Jin oral liquid on downgrade the relative expression of IL-6 and TNF-α.With the late drug intervention group compared,the effect of early intervention was more obvious.Conclusion:(1)Intrauterine infection can lead to premature brain injury rat and the neuroethology changes,and the relative expression quantity of IL-6 and TNF-α increase in the brain tissue.(2)Pu Jin oral liquid can improve neurobehavioral ability of early litter rat with brain damage,and early intervention effect is superior to the late intervention.(3)Pu Jin oral liquid can reduce relative expression of IL-6 and TNF-α of the premature rats caused by intrauterine infection in the brain tissue,and the effect was more apparent of early drug intervention. |
