Objective:(1)To observe the effects of Pu Jin oral liquid on the brain pathology,neuroethology,expression of Nogo-A m RNA and OMgp m RNA in the brain damage rats caused by intrauterine infection/inflammation.(2)To compare the different effcts among the groups of PuJin oral liquid of high and low dose and ginkgo biloba extract,late and early intervention.?To discuss "phlegm" and "blood stasis" is the main pathological basis of premature infant brain injury,and to provide experimental evidence for early application of the Chinese medicine of promoting blood circulation and eliminating phlegm for resuscitation.Methods:(1)30 pregnant rats were randomly divided into lipopolysaccharide(LPS)group(n = 24)and the control group(n = 6),injected respectively LPS with 350μg/kg·d and physiological saline on the first 16 th,17th days of pregnancy.we called pups which birthed less than 22 days pregnancy as the premature rats.(2)80 newborn premature rats of 0-day-old were randomly divided into eight groups-early drug intervention groups(A1、B1、C1、D1)and late intervention groups(A2、B2、C2、D2).A1 and A2 are Pu Jin oral liquid of low dose groups,B1 and B2 are Pu Jin oral liquid of higher dose groups.C1 and C2 are ginkgo biloba extract groups.D1 and D2 are model groups.(3)The early intervention group rats were started to receive gastric perfusion from 0-day-old and the late intervention group rats received gastric perfusion from 7-days-old.The gastric perfusion were both last for 7 days.After the intervention,all the rats breed to 21-days-old with the same conditions.(4)The Production situation of pregnant rats and the birth weight,color,reaction ability and activity of newborn rats and the neuroethology score were observed at newborn and 21-days-old respectively.The intrauterine infection/inflammation status of uterine and placenta and the brain pathology of newborn rats were observed after rates borning by HE staining and RT-PCR method detect the expression of Nogo-A m RNA and OMgp m RNA extracted from the brain tissue of newborn and 21-days-old rats.Results: ?General situation: The pregnant rats of LPS group after modeling were easier to abortion and premature birth,even Died in childbirth.Compared with the control group,pregnant rats of LPS group give birth to less newborn rats(P < 0.05).And the early neonatal rats of LPS group have low weight(P<0.05),less activity,skin color,violet purple and weak stress reaction.?Many neutrophil infiltration and vascular congestion are found on the uterus and placenta pathology of pregnant rat in the LPS group.And there is not discovering any inflammatory reaction in the the control group.From the brain pathological of early neonatal rats in the LPS group,we find that the structure of the white matter、 nternal capsule and corpus callosum are sparse,and nerve fibers arrangement messily,the number of neurons is less.?Compared with the model group,Pu Jin oral liquid high and low dose and ginkgo biloba extracts all can improve neurobehavioral ability of the early neonatal brain damage rats(P<0.05),and the effect of early intervention is superior to the late intervention(P<0.05).But there were no significant difference among Pu Jin oral liquid high and low dose and ginkgo biloba extract on improving neurobehavioral ability of the early neonatal brain damage rats(P>0.05).(4)On the relative expression of Nogo-A m RNA and OMgp m RNA extracted from brain tissue of newborn rats,the control group and the LPS group were significantly different(P<0.05).Both the early intervention groups and the late intervention groups,compared with model group,Pu Jin oral liquid high and low dose and ginkgo biloba extract could significantly reduce the relative expression of Nogo-A m RNA and OMgp m RNA(P<0.05).In the early intervention group,Pu Jin oral liquid high dose could reduce relative expression of Nogo-A m RNA and OMgp m RNA obviously compared with the low dose and ginkgo biloba extract(P<0.05).But there was no significant difference between Pu Jin oral liquid low dose and ginkgo biloba extract(P>0.05).In the late intervention group,Pu Jin oral liquid high dose could reduce relative expression of Nogo-A m RNA and OMgp m RNA obviously compared with the low dose and ginkgo biloba extract(P<0.05).And there was significant difference between Pu Jin oral liquid low dose and ginkgo biloba extract(P<0.05).The effect on decreasing Nogo-A m RNA and OMgp m RNA relative expression between the late intervention groups and the early intervention groups were significant difference(P<0.05).Conlusion:?Intrauterine infection/inflammation can lead to the neuroethol ogy of early neonatal brain damage rats transform and brain tissue increase.?Pu Jin oral liquid can decrease the relative expression of Nogo-A m RNA and OMgp m RNA extracted from brain tissue of early neonatal brain damage rats,and the effect of early intervention is better than late intervention.It can pro vide experimental basis for early application of treating the traditional Chinese medicine of promoting blood circulation and eliminating phlegm for resuscitation on treating premature brain damage induced by intrauterine infection/inflammat ion.?We confirmed that the pathological mechanism of traditional Chinese m edicine of premature brain damage induced by intrauterine infection/inflammati on is mainly the theory of "phlegm" and "blood stasis". |
