Objective :The study aims to verify the method of building rat models of bronchiolitis obliterans,and provide experimental basis for further study of its pathological mechanism;to understand the effects of the drugs on type I collagen(ColⅠ)and type III collagen(ColⅢ)deposition in lung tissue of rat models,and furthermore,to explore new methods for the treatment of BO.Methods: 36 rats were randomly divided into two groups: group A--control group(n = 12)and group B--model group(n = 24),and rats in group B were instilled with diacetyl through trachea at a dosage of 125 mg per kilogram and rats in group A were dealt in the same method and same dosage,however,distilled water was used instead of diacetyl.After 7 days,6 rats of both two groups were chose randomly and signed as group A1 and group B1;these 12 rats were executed and their lung tissues were taken out.,which were then dealt with HE staining to observe the pathological change,and judge whether the model is successfully built.Remaining 6 rats in group A were signed as group A2 and remaining 18 rats in group B were randomly divided into three subgroups and signed as group B2,group B3,group B4,respectively,which,along with group A2,formed 4 groups in the second part of the experiment.Rats in these 4 groups were dealt with different medicine:rats in group A2 and B2 were given gavage with normal saline while those in group B3 and B4 were given gavage with Wenyang Tongbi Tang and prednisone,respectively.And gavage was given one time per day for continuous 21 days.On the next day of last intragastric administration,pulmonary function of all these rats were tested and then,their blood and lung tissue samples were collected,which were then made slice,HE staining was used to observe the changes of the lung structure in each group;immunohistochemical reagent box of Col I and Col III were used to examine the contents of them,photos of each slice were got with Olympus image collecting system and analyzed with Image Pro Plus image analysis system,and the average optical density of each sample was measured;all the data were analyzed with the software SPSS17.0 statistics system.Results: 7 days after modeling,bronchiolitis obliterans-like pathological changes can be seen in rats in group B1,which includes thickened mucosae around small airway and constrictive lumens and some epithelial cells were lost from the wall of the lumen and there were lots of inflammatory cells around the airway and the alveolus pulmonis;while lungs of rats in group A1 were almost normal;After giving drugs for 21 days,rats in group B3 and B4 had a slighter inflammatory infiltration and fibrosis in lung tissues compared to rats in group B2,and the degree of luminal occlusion were also limited;comparing the average optical density of collagen deposition,rats in group B4 has the least ColⅠ in lung tissues than other groups,but unfortunately,the difference among them has no statistic significance(P> 0.05);while rats in group B3 has the least collagen Ⅲ in their lungs than other groups and the difference was statistically significant(P < 0.01).Conclusion Intratracheal instillation of DA provides a feasible modeling method,which could be applied to the experimental research of BO as as a new method of building models;some indexes indicating inflammation could go on while those indicating function of small airway might go down;prednisone might be effective in reducing the content of ColⅠ;both Chinese traditional medicine and western medicine can reduce the deposition of collagen Ⅲ,but Chinese traditional medicine has a better effect.
