Object:This study investigates the expression of NGF, BDNF and NT-3 in the stress conditions of different intensity aerobic exercise. The aerobic exercise program model was proposed by Shu-zhe Ding, with reference to the exercise load standards of Bedford and Sun Xiao-juan.The experiment takes the rats right limb type Ⅱ muscle fibers (gastrocnemius) and type Ⅰ muscle fibers (soleus), and take the central striatum, using immunoblotting to detect the content of NGF, BDNF and NT-3 neurotrophic factor protein. This study explores the peripheral of aerobic exercise (different types of muscle fibers) and central nervous trophic factor protein expressionimpact for sports and fitness, and scientific training.Methods:6 week old male SD rats of 30, stratified according to body weight were randomly divided into normal control group (NC, n= 10), moderate-intensity aerobic exercise group (ME, n=10), high-intensity aerobic exercise group (with HE, n= 10). ME group in which a slope of 5 °19.3m·min-1×60min (equivalent to 76% VO2max); by HE group gradient of 10°,26.8m·min-1×40min (equivalent to 90% VO2max).The rats were killed at anesthesia at the end of the experimental program. The rat brain striatum was takeen in accordance with the stereotactic atlas and the right limb gastrocnemius and soleus wrer taken into liquid nitrogen. This study uses western blot to detect the expression of NGF, BDNF and NT-3 protein.Results:(1) Compared with group NC, the content NGF, BDNF and NT-3 protein of the gastrocnemius and soleus muscles in ME and HE groups increased, NGF/Beta-actin, BDNF/Beta-actin and NT-3/Beta-actin ratio were also increased, there have a very significant difference (p<0.01). Compared with group ME, the content NGF, BDNF and NT-3 protein of the gastrocnemius and soleus muscles in ME and HE groups increased significantly, NGF/Beta-actin, BDNF/Beta-actin and NT-3/Beta-actin ratio increased significantly, there have a very significant difference (p<0.01). The expression of NGF, NT-3 protein in muscle fiber type Ⅰ (soleus) dominated than type Ⅱ muscle fibers (gastrocnemius); and the expression of BNDF was similar to the expression in the gastrocnemius and soleus muscles, there were no significant difference.(2) Compared with group NC, the content of NGF, BDNF and NT-3 protein of striatum in group ME and group HE, the NGF/Beta-actin, BDNF/Beta-actin and NT-3/Beta-actin ratio increases, there have a very significant difference (p<0.01). Compared with group ME, the content of NGF, BDNF and NT-3 protein of striatum in HE group, the NGF/Beta-actin, BDNF/Beta-actin and NT-3/Beta-actin ratio increased significantly, with a very significant difference (p<0.01). The protein of NGF and NT-3 were expressed in the striatum in groupNC, group ME and group HE. The expression of NGF and NT-3 protein of the striatum were lower than the gastrocnemius and soleus muscle. The expression of BDNF protein was on difference between the groups in the gastrocnemius, soleus and the striatum,Conclusions:(1) 8 weeks of moderate-intensity aerobic exercise, and 8 weeks of high-intensity aerobic exercise could induce the expression of NGF, BDNF and NT-3 protein of gastrocnemius, soleus and striatum. The expression of the factors is the most significant in 8 weeks of high-intensity aerobic exercise. (2) The expression of NGF and NT-3 protein in muscle fiber type Ⅰ (soleus) than type Ⅱ muscle fibers (gastrocnemius). The expression of NGF and NT-3 protein in striatum were lower than the gastrocnemius and soleus muscle. The expression of BDNF protein was similar in gastrocnemius, soleus and striatum.
