Objectives:With a mice model of POI based on environmental chemistry VCD,the effects of the Modified Guilu Erxian Decoction on POI mice model of estrus cycle,serum AMH,ovarian tissue morphology and expression of mi R-190 were investigated.The intervention effect and mechanism of the Modified Guilu Erxian Decoction on POI were researched,which can provide a theoretical basis for clinical treatment of POI.Methods:40 female Kunming mice with continuous twice normal estrus cycles were involved in the experiments.They were randomly divided into blank group,model group,DHEA group and traditional Chinese medicine(TCM)group.The mice of the model group,the DHEA group and the TCM group were intraperitoneally injected with VCD for 15 consecutive days to establish the mice model of POI.Then,the mice of the TCM group were given 0.3ml Modified Guilu Erxian Decoction to filled into the stomach,while the DHEA group were given 0.3ml DHEA,and the blank group and the model group were given 0.3ml normal saline.The total treatment was 30-dyas,and 4-days was a period of treatment,stopping 1-day after every 4-days.The serum AMH levels of mice were detected by ELISA after treatment,as well as the estrus cycles and morphology changes of ovarian tissue under light microscope.Furthermore,the q RT-PCR were performed to detect the mi R-190 expression in mice’s ovarian tissue.Results:1.The estrus cycles of mice were changed.The vaginal smears of the blank group mice showed regular estrus cycles.However,the mice in other three groups injected with VCD intraperitoneally had estrus cycles dysfunction,which mainly happened during diestrus.The disordered estrus in the DHEA group and the TCM group had a slight improvement after drug intervention,which showed a extended estrus cycle and diestrus.2.The morphology and histology of mice’s ovarian were changed.The ovaries of the model group showed a apparent atrophy under light microscope and a thickened cortex.Many atretie follicles could be seen,while the primordial follicles and each stage growing follicles disappeared or just were seen occasionally.Compared with the model group,the primordial follicles and secondary follicles of the DHEA group and the TCM group were slightly increased,however the atretie follicles were decreased.3.The serum AMH level of the model group was lower than that of the blank group,but it’s not obvious.The DHEA group and the TCM group had the trend of increasing the serum AMH level compared with the model group,but it’s not obvious.4.The ovarian mi R-190 expression of the model group was higher than that of the blank group,and there was a statistically significant difference(P<0.01).The ovarian mi R-190 expression of the DHEA group and the TCM group were lower than that of the model group,and there was a statistically significant difference(P<0.01).And the ovarian mi R-190 expression of the TCM group was slightly lower than that of the DHEA group,and there was a statistically significant difference(P<0.01).The ovarian mi R-190 expression of the model group was slightly higher than that of the blank group,and there was a statistically significant difference(P>0.05).Conclusions:1.The mice model of POI could be successfully established by using environmental chemistry VCD.2.The Modified Guilu Erxian Decoction could effectively increase the primordial follicles and secondary follicles in POI model mice’s ovarian tissue,the the mechanism may be related to the inhibition of ovarian mi R-190 expression.
