Background and ObjectiveThe development of medical produces many miracles which shock the world. The first test-tube baby in the world was born in the 20 th century which made the researchers see some breaking conventional breeding methods. These methods made the fertility problems of a tens of thousands of infertile patients be solved.Infertility is more common not only in abroad, in China, many couples are suffering from birth difficult. In China, each family have a desire of birthing a healthy baby.The development of assisted reproductive technology bringsbeautifulhope to infertile patients. To some extent, not only maintain the family harmony, but also provideprotection to social harmony. There are more than 30 years of development of ART technology, but the success rate remains to be further improved.In normal female menstrual cycle, embryo implantation finished in a very short time.Endometrial receptivity refers to the receptivity of endometrium to embryo. Implantation window occurs in a woman's menstrual cycle after ovulation 6-7days. At this time, the receptivity of endometrium is the highest and the embryo can implant successfully. Broad scholars have already demonstrated thatleukemia inhibitory factor(LIF) and insulin-like growth factor(IGF) can act on the endometrium and embryo at the same time.They achieved the peak during implantation window period. They were closely relating to the formation of the receptivity of endometrium.In clinical work,older patients with ovarian reserve function to drop lead to the ideal egg quality than younger patients. After the retrieval, the low quality of embryos lead to the low-level embryo growth rate and clinical pregnancy rate after transplanting into the uterine cavity. Many scholars are building the co-culture system to improve the embryo growth rate and clinical pregnancy rate. So far, using endometrium as a breeding layer and building the co-culture system with embryos is studied maturely. Because the co-culture system is closed to the mother's growth environment, it can provide all kinds of embryonic growth material, at the same time, it can secrete a variety ofrelated factors which can help embryo growth and improve the receptivity of endometrium, such as LIF, IGF, EGF, and other factors.To a certain extent, the co-culture system can improve the activity of embryo, help to promote the growth of embryo, and further improve the embryo implantation rate and clinical pregnancy rate. In this background, this study selects endometrium and embryo to build the co-culture system and uses the RT-PCR method to detect the expression level of LIF and IGF in the lining of the uterus and to explore the expression rule of the implantation window to improvethe effect and possible mechanism of the receptivity of endometrium. At the same time, this study observes the growth situation of trained under the condition of the embryo, discusses the superiority of the co-culture system, further provides theoretical basis for optimization of clinical treatment. Materials and Methods Choose 10 patients from the second affiliated hospital of zhengzhou university artificial assisted reproductive center in June 2014 to June 2015, they all cancel the transplant after taking eggs because of OHSS. Taking the endometrium after the eggs be taked 3 days, and building the co-culture group with laboratory embryos(group A), and using the single endometrium group(group B) and the single embryo group(group C) as control group.By RT-PCR methoddetectingthe expression level of LIFmRNA and IGFmRNA in the co-culture group(group A) and the single endometrium group(group B)in D3, D4, D5, D6, D7 day, and using a microscope toobserve the embryo growth in the co-culture group(group A) and the single embryo group(group C).The SPSS20.0 data processing software is used, the data obtained by repeated measurement data analysis of variance, P < 0.05, said the difference was statistically significant. Results1.LIF and IGF mRNA show positive expression in the co-culture group(group A) andexpress quantity increased in D3, D4, D5, D6, D7 day, in D6, D7 day show strong positive expression, the expression quantity achieve to the peak.2.LIF and IGF mRNA show positive expression in the single endometrium group(group B)andexpress quantity increased in D3, D4, D5, D6, D7 day, in D6, D7 day show strong positive expression, the expression quantity achieve to the peak.3.LIF and IGF mRNA express inthe co-culture group(group A) are significantly higher than in the single endometrium group(group B), and the difference was statistically significant(P < 0.05)4. Observe the embryo growth under a microscope can find that the co-culture group(group A) has less embryo pieces, less chance of uniform, and the development speed of embryo is relatively quicker than the single embryo group, and the proficiency of morula and blastocyst are higher than the single embryo group. Conclusions1.LIF and IGF in endometrium is the factor of the receptivity of endometrium.2.The D6,7 days after ovulation is the implantation window, at this point the receptivity of endometrium is the best, the highest ability to accept embryo is the best, and the highest ability of embryo implantation is also the best.3.The co-culture system is more accessing the mother's growth environment. The endometrium can secret many cell factors which can help the embryos implantation. The co-culture system promotes interaction between endometrium and embryo and helps mutual development.
