Pulvis Glycyrrhizae Praeparatus is special processed by Leguminosae l icorice.The character and taste of Pulvis Glycyrrhizae Praeparatus is sw eet,salty and cold with the effect of heat-clearing,cooling blood and p urging fire detoxification.At home and abroad,the Licorice reports mor e than Pulvis Glycyrrhizae Praeparatus about material base.Pulvis Glycyr rhizae Praeparatus is belonging to the clearing away heat and toxic drug s which has the effect of Antibacterial,anti-viral and anti-inflammatory.According to ancient records,the therapeutic effect of Pulvis Glycyrrhiz ae Praeparatus to the plague is quite remarkable.Because of the unique processing method of it,out of the safety and hygiene considerations,t here is a trend of Pulvis Glycyrrhizae Praeparatus having been eliminate d.In order to explore the great treasure of Chinese medicine,discarding the dross and selecting the essential,there is a need for a more syste matic study about it.Setting up the quality standard of it in this paper,the characters,identification,inspection,fingerprints and content determi nation of it were studied,the antibacterial and anti-influenza virus were studied in vitro.The micro identification method was used to observe the microscopic characteristics of it,which found that the characteristics of the powder were similar to that of the microscopic characteristics of licoricea,and two batches of them were mixed with false medicinal herbs;The results of the TLC identification was ideal,which was based on the control of the licorice,licorice and licorice;According to the determination of moisture content(2015 China Pharmacopoeia four general rules 0832 second method(drying method))measure water content in ten batches of P ulvis Glycyrrhizae Praeparatus,the results showed that the water content of the samples were less than 9%;The method of Pulvis Glycyrrhizae Praeparatus’ microbial limit test was validated.According to the determi nation of Control bacterias(2015 China Pharmacopoeia fourgeneral rules 1106),Escherichia coli and Bile salt resistant gram negativebacteria were not checked in the three batches of Pulvis Glycyrrhizae Praeparatus.According to Requirements of TCM injection fingerprints techniques p romulgated by China Food and Drug Administration(CFDA),the chrom-atographic fingerprints of Pulvis Glycyrrhizae Praeparatus were establish ed by HPLC method.Seventeen common peaks in chromatographic finger prints were identified,All common peaks came out in 90 minutes and th e resolution was high.Ten batches of Pulvis Glycyrrhizae Praeparatus we re compared with the HPLC-reference chromatogram,the similarity was a ll up to 0.9.Another two batches of samples were detected wuich was o bvious compared with the HPLC-reference chromatogram.Microscopic ide ntification was proved to be counterfeit goods.The results of the two me thods’ identification were consistent.HPLC method was established to determine the content of Pulvis Gly-cyrrhizae Praeparatus by useing the index of liquiritin,liquiritigenin and glycyrrhizic acid.Results: the peak separation of all were good.Liquiritin,liquiritigenin and glycyrrhizic acid has a good linear relationship in the irrespective concentration ranges.The recovering rate was between 97.49% and 103.24%.RSD was between 0.58% and 1.24%.Conclusion: the method is accurate,simple and reproducible,which can be used for the quantitative study of licorice,licorice and licorice acid in Pulvis Glycyrrhizae Praeparatus.Pulvis Glycyrrhizae Praeparatus and licorice had anti-bacterial effects on Escherichia coli,Pseudomonas aeruginosa,Staphylococcus aureus and C andida albicans to some extent by useing of the paper diffusion method and double dilution methods.There were significant anti-bacterial and bac tericidal effects on Staphylococcus aureus in both the Pulvis Glycyrrhiza e Praeparatus and the licorice.MIC was 0.016g/ml,MBC was 0.031g/ml.Theinhibitory effect of Pseudomonas aeruginosa and Escherichia coli was weak in both the Pulvis Glycyrrhizae Praeparatus and the licorice.In ad dition,the MIC of Pulvis Glycyrrhizae Praeparatus was 0.250g/ml to Ca ndida albicans,but the MIC of licorice was more than 0.500g/ml to Ca ndida albicans.That was to say the effect of inhibiting Candida albicans Candida albicans was stronger than that of licorice.MTT method was used to study the anti H1N1 activity of the different polar extracts from Pulvis Glycyrrhizae Praeparatus in vitro.The results showed that total extract of Pulvis Glycyrrhizae Praeparatus,total extract of licorice,ethyl acetate(Et OAc)extract,n-butyl alcohol(nBu OH)extrac tand the Ribavirin group(positive control group)had the effect of anti-i nfluenza virus biosynthesis,the therapeutic index(TI)were 5.16±0.24、2.41±0.11、7.54±0.38、49.52±2.31、6.88±0.36;Water extract is the str-ongest part of anti-influenza virus biosynthesis,however total extract of licorice is the weakest of that,the effect of anti-influenza virus biosynt hesis in water extract and n-butyl alcohol extract was better than the Ri bavirin group;The n-butyl alcohol(nBuOH)extract kill the virus directl y,the therapeutic index(TI)was 3.34±0.37.There was no significant diff errence between Ribavirin group and n-butyl alcohol extract on killing t hevirus directly,the therapeutic index(TI)was 3.91±0.34,however total extract of Pulvis Glycyrrhizae Praeparatus,total extract of licorice,the water extract has no direct effect of killing virus;different extract of P ulvis Glycyrrhizae Praeparatus,total extract of licorice,Ribavirin group w ith different concentrations had the cytopathogenic effect of typical influ enza virus induced,which has no significant difference with Virus contr ol,that was to say all these drugs had no effect of interdicting virus ads orption.
