Lignum Pini Nodi is the valuable medicinal product in China. The branch with the node or bend can be soaked and taken for rheumatic condition, analgesic and anti-cancer. The Chinese pine (Pinus tabulaeformis carr.) and Masson pine (Pinus massoniana Lamb.) are the common available resource of Lignum Pini Nodi. However, the relevant quality standard hasn't been established yet. Therefore, systematic research works have been done in this paper.By researching the discrimination method, content determination method and standard formulation of the items, the standard of Lignum Pini Nodi was established. The results were shown as follow:1. Based on the textual research on historical herb literatures and major herb markets investigation, the common resource of Lignum Pini Nodi from the node or bend of Chinese pine or Masson pine has been clarified.2. Based on the direct observation of the shape, cross section and powder microstructure, the character of Lignum Pini Nodi has been confirmed. On the other hand, the TLC analysis with the indicativeα-terpineol has been done. A physical and chemical standard has been established for the quick identification of Lignum Pini Nodi.3. The effect of powder size and extraction time on the total volatile oil recovery has been studied. The results indicated that the powder size designed for the test can not affect the extract efficiency of total volatile oil. After 8 h extraction, most of volatile oil in Lignum Pini Nodi will be totally recovered.4. The factors effecting on the extraction ofα-pinene has been studied and optimized. The different extraction method, extraction solvent and extraction time have been designed for screening the optimum process. The results showed that theα-pinene in Lignum Pini Nodi can be totally extracted under the condition of ultrasonic extraction process with ethanol (1:10, solid to solvent) during 15 min. The content ofα-pinene has been determined by the GC with hydrogen flame detector. A good linear correlation can be observed,α-pinene show a good linear relationship in the range of 0.01 -10.0 mg/mL(r = 0.9999). The average recovery rate is 99.7%.5. The quality control protocol of Lignum Pini Nodi for Chinese Pharmacopea (edition 2010) was drafted. The cotent of total volatile oil was no less than 1.00%, and the content ofα-pinene was no less than 0.35%.6. The fingerprint method of Lignum Pini Nodi was established, and the comparison of the different species and areas has been done in this paper.According to the ChP2005, the processing of the Radix Et Rhizoma Glycyrrhizae was identified as follows: removing the impurity, cleaning, soaking, cutting into slice, drying. However, there was no relavant regulation on the specific mothod for soaking and quality standard decoction pieces. Therefore, relavant works have been done in this paper.1. Based on the direct observation of the shape and powder microstructure, the character of Radix Et Rhizoma Glycyrrhizae Decoction Pieces has been confirmed. On the other hand, the TLC analysis has been done.2. The contents and stability of liquiritin and glycyrrhizic acid have been determined, using the methods of ChP2005.3. The stability of Radix Et Rhizoma Glycyrrhizae Decoction Pieces was tested, and the results show that the quality of the pieces was stable.4. The fingerprint method of Radix Et Rhizoma Glycyrrhizae was established, and the results show no significant differences between Radix Et Rhizoma Glycyrrhizae and its decoction pieces.
