| ObjectiveWe established chronic atrophic gastritis(CAG)models,the type of deficiency of the spleen and stomach in rats,from the rat gastric emptying rate,serum gastrin(GAS),the activity of pepsin,vascular endothelial growth factor(VEGF)expression and multi aspects research.Then,we explored Jie Mei Zhang professor my mentor in clinical common modified Xiangsha Liujunzi decoction and its reduction mechanism on CAG rats with deficiency spleen and stomach,and provided experimental support for clinical application of this method to treat the disease and to help in the development of traditional Chinese medicine of new products,to provide new ideas and methods.MethodsWe selected 76 SPF grade healthy Wistar rats,male and female,in the feeding environment barrier in SPF.The first to be fed for 7 days,then were randomly divided into blank control group(K),male and female each 6,CAG model group,male and female each 32.Rats in the blank control group were free to eat and drink.The model group was given 20mmol/L deoxycholic acid sodium free to drink,and the model group was made for 1 months.During every 10 days by gavage once 60% alcohol,2ml/only;after January,with 10mmol/L deoxycholic acid sodium and 30% alcohol weekly alternating free drinking,hunger and give adequate feeding,feeding 2 days,fasting for 1 days.The model rats died 20 after 100 days of modeling.Then,we randomly choosed the blank control group 1 rat of male and female,model group of 2.They were sacrificed to observe the model duplication,until after the success of the model,the successful model of the 40 rats were randomly divided into model group(M),Weifuchun group(W),Folic Acid Tablets group(Y),and the treatment group(Z),with 10 rats in each group.K group and M group gave saline 10ml/kg gavage.W group gave Weifuchun 0.45g/Kg gavage,formulated as 0.045g/ml the standby liquid,Y group gave folic acid tablets 2mg/kg irrigation stomach,0.2mg/ml suspension liquid reserve and Z group gave modified Xiangsha Liujunzi Decoction 10g/Kg gavage,fried and then concentrated to 1g/ml of liquid reserve.Drugs intervened one month,then drew blood and materials.We calculated the Gastric emptying rate,the activity of pepsin,the level of serum gastrin(GAS)and vascular endothelial growth factor(VEGF)expression.Results1.Through this experiment,we successfully established a rat model of chronic atrophic gastritis of spleen stomach deficiency by a combined method of modeling.The experiment was carried out in 76 rats.12 rats were in the blank control group,2 of which were killed.64 models were executed,4 of which were killed and 20 died.The successful rate was 68.75%.2.Gastric emptying rate:compared with the blank control group,model group rats gastric emptying rate decreased significantly(P<0.01);compared with the model group,treatment group,folic acid tablets group and Weifuchun group could significantly increase the rat gastric emptying rate.The treatment group and Weifuchun group are better(P<0.05).3.The activity of pepsin comparison;compared with the blank control group,pepsin activity of rats in model group decreased significantly(P<0.01);compared with the model group,treatment group,folic acid tablets group and Weifuchun group can enhance the pepsin activity of rats.The treatment group and Weifuchun group are better(P<0.05).4.The level of serum gastrin : compared with the blank control group,rats in the model group,the serum gastrin increased significantly(P<0.01);compared with the model group,treatment group,folic acid tablets group and Weifuchun group can reduce the secretion of rat serum gastrin,the effect is obvious(P<0.05).5.VEGF expression: compared with the blank control group,VEGF of rats in the model group showed strong positive expression(P<0.01);compared with the model group,VEGF in treatment group,folic acid tablets group and Weifuchun group showed weak positive expression(P<0.05).The treatment group is better(P<0.05).Conclusion1.Modified Xiangsha Liujunzi Decoction of experimental study and clinical study were achieved with satisfactory results.It can be further modified and applied in clinical practice.2.Modified Xiangsha Liujunzi Decoction can improve the rat gastric emptying rate,pepsin activity,reduce the secretion of gastrin and reduce VEGF expression.Its treatment of CAG may be connected with promoting gastrointestinal function,protecting gastric mucosa,promoting its repairing,inhibiting the excessive proliferation of vascular endothelium cell and reversing the atrophy.In clinic,it can also reverse the role of intestinal metaplasia and so on. |
